摘 要:为了探明猪粪堆肥升温期的细菌多样性和堆肥过程中由"梯度效应"导致的细菌空间分布差异,采集了堆体上、中、下3层的堆肥样品,对其进行了16S rDNA序列和PCR-DGGE分析。序列分析结果表明:从3层样品中获得的121条16S rDNA序列,其中的113条与GenBank数据库中已知菌的16S rDNA序列表现出了同源性,且大多数与梭菌属有较近的亲缘关系,7条序列与已知菌的序列无任何相似性。上层样品中Shigellasp.、Acinetobactersp.和Clostridiumsp.占优势;中层样品中Clostridiumsp.占绝对优势;下层样品中Clostridiumsp.和Lactobacillussp.占优势。PCR-DGGE结果显示:3层堆肥样品都具有较丰富的微生物种群多样性,同时存在着明显的优势种群结构变化,中层多样性最为丰富。To investigate the bacterial diversity of swine manure composts in the initial stage and the different spatial distribution due to gradient effect,samples in the superstratum,middle-level and substrate of a composting pile were taken and analysed using 16S rDNA and PCR-DGGE technology.The resuls of 16S rDNA sequences showed that 113 out of the obtained 121 stems of 16S rDNA sequences from the three samples displayed homology with those reported in the GenBank database,and most of them related to Clostridium sp.,other seven 16S rDNA sequences were not closely related to any known species.The bacterium related to Shigella sp.,Acinetobacter sp.and Clostridium sp.were dominated in the superstratum sample,the microbe related to Clostridium sp.were absolute dominant position in the middle-level sample,and the bacterium related to Clostridium sp.and Lactobacillus sp.were dominated in the substrate sample.PCR-DGGE profile displayed that the three samples possessed relatively abundant bacterial diversity,especially the middle-level sample,and presented changes of the dominant species among the three samples.
