摘 要:目的:优选枳壳中脂溶性部位成分的最佳提取工艺。方法:HPLC测定枳壳中5种脂溶性成分meranzin hydrate、马尔敏、川陈皮素、红橘素和葡萄内酯的含量,以5种脂溶性成分的含量和脂溶性部位浸膏得率为指标,采用L9(34)正交试验法优化枳壳脂溶性部位成分的提取工艺。结果:枳壳优化后的脂溶性部位成分提取工艺为A2B2C2D2,乙醇浓度85%、料液比1∶8、水浴回流1h,提取2次;以5种脂溶性成分的含量计算,则脂溶性部位成分平均含量为2.86(mg/g),RSD为1.32%,脂溶性部位浸膏的平均得率为2.81%,RSD为1.78%。结论:该提取工艺方法简单、稳定可行,可为枳壳中脂溶性部位成分的大规模化生产工艺提供理论依据。AIM: To study the optimum extraction of liposoluble constituents from immature bitter orange(Fructus Aurantii). METHODS: The content of meranzin hydrate,marmin,nobiletin,tangeretin,auraptene in immature bitter orange were determined by RP-HPLC and the extraction yield was adopted as marker,the orthogonal design L9(34) was used to evaluate extraction. RESULTS: The optimal parameters were as follows: immature bitter orange extract was produced with 70% ethanol refluxing,1 h,2 times,herb-to-extract ratio was about 1∶ 8. The mean content of five constituents was 2. 86(mg/g)( RSD = 1. 32% ) and the extraction yield of liposoluble constitutents was 2. 81% ( RSD = 1. 78% ). CONCLUSION: This method is simple,stable,reliable and can provide solid basis for large scale extraction of liposoluble constitutents.
