摘　　要：目的观察葛根素对脂多糖(LPS)诱导小鼠RAW264.7巨噬细胞表达诱导型一氧化氮合酶(i NOS)的影响。方法用脂多糖刺激小鼠巨噬细胞RAW264.7,与不同浓度的葛根素进行培养,采用逆转录多聚酶链式反应(RT-PCR)和免疫蛋白印迹法测定(Western blotting)方法分别检测RAW264.7巨噬细胞i NOS mRNA和蛋白质表达。结果随着葛根素的浓度的增加,RAW264.7细胞i NOS的mRNA及其蛋白质表达逐渐减少(P<0.01)。结论葛根素可以通过调节RAW264.7细胞表达i NOS发挥抗炎的作用。Objective To investigate the effect of Puerarin on expression of inductible nitric oxide synthase（iNOS）in RAW264.7 macrophage cells induced by lipopolysaccharide（LPS）.MethodsThe protein and mRNA levels of lipopolysaccharide（LPS）-induced iNOS was determined in RAW264.7 macrophage cells using Western blotting and RT-PCR respectively.ResultsPuerarin inhibited expression of iNOS concentration dependently（P0.01）.ConclusionPuerarin may reduce iNOS level,result in prevention of inflammation.
