摘  要:茄尼醇在生物化学中有着非常重要的作用,主要用于合成辅酶Q10、维生素K2、抗癌增效剂SDB.主要研究探索提取废次烟草中茄尼醇的适宜条件.通过采用两相提取与超声波辅助的方式从废弃烟叶中提取茄尼醇,经皂化得到茄尼醇粗品,并通过大孔吸附树脂进一步纯化,得到纯度较高的茄尼醇.讨论了影响提取率的主要因素,确定了优化的工艺参数:用90%(体积分数)甲醇与石油醚体积比为1∶1作提取剂,料液体积比1∶4,超声波浸提3 h,得到茄尼醇纯度是27.2%(质量分数),提取率为88.5%,经皂化大孔吸附树脂纯化得茄尼醇质量分数达90%以上.为茄尼醇的提取与浓缩提供了一种新的方法.Solanesol plays a very important role in biochemistry,mainly in the field of synthesizing coenzyme Q10,vitamin K2 and SDB.It belongs to polyisoprene alcohol and displays strong ability to eliminate free radicals,excellent bioactivities such as anti-oxidation,anti-radiation,anti-inflammation,and fungicidal activity for its nine non-conjugated double bonds in the structure.It has been widely used in production of healthy products,cosmetics,anti-hypertension medicines,and anti-tumor drugs,with high market prospects.The extraction and purification of solanesol from discarded tobacco leaves were studied.Through ultrasound-assisted two-immiscible organic solvents extraction and saponificaion,semi-finished solanesol was obtained.Then purified by macroporous adsorbing resins,the purity of solanesol can be reached a high level.The main factors on extraction yield of solanesol were optimized by orthogonal design,the optimum extraction conditions were investigated as follows:with ultrasound-assisted time 3 h,two-immiscible organic solvents 90% methanol and petroleum ether(1∶1,by vol.) as solanesol extraction solvent(V(tobacco)∶V(solvent)=1∶4),which simultaneously separate solanesol form series of polar impurities,solanesol can be obtained with the purity of 27.2% and the yield of 88.5%,which was higher than those reported in literatures.The purity of solanesol can be reached 90% after saponification and macroporous adsorbing process.This technology could provide a new,simple extracting procedure of solanesol from discarded tobacco leaves.
