摘 要:目的观察体外培养时灯盏花素对大鼠腹膜间皮细胞生长和增殖的影响,探讨灯盏花素对间皮细胞的保护机制。方法对SD大鼠腹膜间皮细胞进行分离培养,第三代用于实验,以1×103细胞接种于96孔培养皿,分成8组,每组9孔,分别加入不同浓度的灯盏花素和LPS。培养48 h和72 h后通过细胞计数了解间皮细胞生长情况;培养72 h后加入MTT 5 mg/孔,用酶标仪490 nm比色,了解间皮细胞增殖情况。结果 (1)细胞数量影响:在未加LPS组中,25μL灯盏花素组和12.5μL灯盏花素组间皮细胞生长良好,与单纯培养基组相比差异无显著性(P=0.086、P=1.000);50μL灯盏花素组与单纯培养基组相比差异有显著性(P=0.004)。而加LPS组间皮细胞生长情况差于未加组,在LPS组中,加入灯盏花素各组拮抗LPS对间皮细胞生长抑制作用(P=0.000)。(2)细胞增殖的影响:未加LPS组中单纯培养基组、25μL灯盏花素组和12.5μL灯盏花素组的间皮细胞增殖差异无显著性(P=0.64、P=0.929),50μL灯盏花素组的间皮细胞增殖与单纯培养基组比较差异有显著性(P=0.039)。加有LPS组中50μL灯盏花素组、25μL灯盏花素组组间差异无统计学意义(P=0.771),其余各组间差异统计学意义(P=0.000),其中LPS组间皮细胞增殖最差,12.5μL灯盏花素组次之。结论 (1)灯盏花素间皮细胞的生长和增殖具有保护作用。(2)灯盏花素能够减轻LPS引起的免疫炎症造成的间皮细胞损伤,且在一定浓度范围内存在量效正相关关系。Objective To study the effects of erigeron on the growth and proliferation of peritoneal mesothelial cells in vitro,and to study its mechanism.Methods Peritoneal mesothelial cells isolated from SD rats were divided into 8 groups according to culture medium: medium only(Group 1),50 μL erigeron(Group 2),25 μL erigeron(Group 3),12.5 μL erigeron(Group 4),50 μL erigeron plus 10 μL LPS(Group 5),25 μL erigeron plus 10 μL LPS(Group 6),12.5 μL erigeron plus 10 μL LPS(Group 7),and 10 μL LPS(Group 8).Cell count(at 48 hour and 72 hour) and MTT(at 72 hour) were used for investigation of the growth and proliferation of peritoneal mesothelial cells.Results(1) There was no significant difference in mesothelial cell count between Group 1 and Group 3 or 4,respectively(P〈0.05).However,mesothelial cell count in Group 5 was significantly higher than that in Group 1(P〈0.05).Meanwhile,cell counts in groups cultured with medium plus LPS were less than that in those cultured without LPS.Furthermore,erigeron showed significant protective effect on cell count reduction induced by LPS(P〈0.001).(2) There was no significant difference in cell proliferation observed with MTT among Group 1,3 and 4(P〉0.05).However,MTT showed cell proliferation in Group 2 was significantly more active than that in Group 1(P〈0.05).Except for between Group 5 and 6,there were significant differences in cell proliferation among Group 5,6,7 and 8(P〈0.001).Moreover,erigeron induced concentration-dependent protective effect on cell proliferation.Conclusion(1) Erigeron has protective effect on mesothelial cells.(2) Erigeron attenuates LPS-induced mesothelial cell injury in a concentration-dependent manner.
