摘 要:为探讨不同来源的促性腺激素对猪卵母细胞早期孤雌发育的影响,将猪卵母细胞置于添加有不同来源促性腺激素(分别来自美国Sigma公司和中国科学院动物研究所)的成熟液中体外成熟40~44 h,成熟卵母细胞经化学孤雌激活后放入胚胎培养液中继续培养,分别于第48小时和第168小时统计卵裂率和囊胚发育率。结果发现,成熟液中同时添加0.5μg/mL中国科学院动物研究所生产的促性腺激素(FSH和LH)的卵裂率和囊胚发育率均略高于其他处理组,分别达到(65.68±24.13)%和(28.41±18.74)%;在囊胚细胞数方面,无论是添加中国科学院动物研究所还是Sigma公司生产的促性腺激素,均以添加浓度为0.5μg/mL时的囊胚细胞数最高。结果表明,两种来源促性腺激素对猪卵母细胞孤雌激活后早期发育的影响无显著差异,说明中国科学院动物研究所生产的促性腺激素(FSH和LH)能有效促进猪卵母细胞体外成熟和进一步孤雌发育效果,且以在成熟液中同时添加0.5μg/mL FSH和0.5μg/mL LH为宜。The present experiment was an attempt to study the effects of different sources of gonadotrophin which were produced by Chinese Academy of Sciences(CAS) and SIGMA of USA on early parthenogenetic development of porcine oocytes in vitro.The porcine oocytes from local abattoir ovaries were matured in maturation medium in vitro for 40-44 hours,and then the mature oocytes were activated parthenogenetically and chemically.The activated oocytes were cultured for seven days in embryo culture medium.The cleavage rate was examined after 48 hours and blastocyst rate was examined after 168 hours.The results indicated that the cleavage rate and the blastocyst rate of the second treatment supplemented with CAS's gonadotrophin(0.5 μg/mL FSH+0.5 μg/mL LH) were a bit higher than those of other treatments,being(65.68±24.13) % and(28.41±18.75) %,respectively.The highest cell number of blastocysts was obtained from the second treatment and the fourth treatment [CAS-and SIGMA-gonadotrophin respectively(0.5 μg/mL FSH+0.5 μg/mL LH)](P0.05).It was concluded that there was no difference between the two sources of gonadotrophin selected in this experiment on early parthenogenetic development of porcine oocytes in vitro.The CAS's gonadotrophin [(0.5 μg/mL FSH+0.5 μg/mL LH) was optimal] could have excellent effects on maturity and parthenogenetic development of porcine oocytes in vitro.
