摘  要:[目的]研究杨梅(Myrica rubraSieb et Zucc.)组织培养过程中防污染和防褐化问题。[方法]以东魁杨梅品种茎段作为离体培养材料,研究不同消毒灭菌方法、不同茎段部位和不同防褐化措施对外植体离体培养的影响。[结果]以75%乙醇1min+0.1%HgCl27min对外植体的灭菌效果最好,污染率为30.00%,褐化枯死率为30.00%,茎段诱导成功率为40.00%;在3个茎段部位中,以半木质化茎段诱导效果最好,污染率为30.00%,褐化枯死率为33.33%,诱导成功率为36.67%;在培养基中添加活性炭1.5g/L+聚乙烯吡咯烷酮(PVP)2.5g/L对外植体防褐化效果最好,褐化率由对照的60.93%下降到21.88%。[结论]该研究可为杨梅优良品种的扩繁推广和高效遗传转化体系的建立奠定良好的基础。[Objective] The research aimed to study the prevention of pollution and browning in tissue culture of Myrica rubra Sieb et Zucc..[Method] Taking the stem segment of Dongkui Myrica rubra Sieb et Zucc.species as the in vitro material,the effects of different disinfection and sterilization,different stem parts,different prevent browning measures to external plants in vitro culture were studied.[Result] The results showed that the sterilizing effect of 75 % alcohol 1 min + 0.1% HgCl2 7 min on the external implant was the best,and the pollution rate was 30.00%,browning and withered rate was 30.00%,stem induction success rate was 40.00%.In the three parts stems,the semi-lignified stem segments was the best,contamination rate was 30.00%,browning and withered rate was 33.33%,success rate was 36.67%.Adding the activated carbon 1.5 g/L + polyvinylpyrrolidone(PVP)2.5 g/L in the medium had the best effect to the external plant anti-browning,and the browning rate from control 60.93% dropped to 21.88%.[Conclusion] The study can lay good basis for extending of good Myrica rubra Sieb et Zucc.species and establishing its effective genetic transformation system.
