摘  要:[目的]研究玉米和胡萝卜体胚发生中的生理生化变化,从而为调控其体胚发生过程提供依据。[方法]研究单子叶植物代表玉米和双子叶植物代表胡萝卜体胚发生的适合培养基,并利用电泳技术对其体胚发生过程中的可溶性蛋白进行比较。[结果]培养基MS+6-BA1.0mg/L+2,4-D0.2mg/L+CH500mg/L较适于玉米愈伤组织的发生;培养基B5+2,4-D0.2mg/L+6-BA0.2mg/L较适于胡萝卜愈伤组织的发生。玉米愈伤组织中存在活跃的蛋白表达;而胡萝卜愈伤组织中的蛋白表达水平则比下胚轴对照明显下降。二者蛋白质表达的差异可能与物种差异有关。[结论]发现了单子叶植物和双子叶植物体胚发生中蛋白质表达的规律。[Objective] The reference for the adjustment and control of somatic embryogenesis of maize and carrot was provided through the research on the physiological and bio-chemical variation of its development.[Method] The medium suitable for the somatic embryogenesis of monocotyledon-maize and dicotyledon-carrot was experimented and the soluble protein during the somatic embryogenesis of maize and carrot was compared based on the electrophoresis technology.[Results] The results showed that the medium:MS+6-BA 1.0 mg/L+ 2,4-D 0.2 mg/L+CH 500 mg/L was suitable for the maize callus-inducing and the medium B5+2,4-D 0.2 mg/L+6-BA 0.2 mg/L was suitable for carrot callus-inducing.There was active protein expression in the maize callus and the protein expression in the carrot callus was not much more active than that in the hypocotyl(CK).[Conclusion] The principle of protein expression during the somatic embryogenesis of monocotyledon and dicotyledon was explored.
