摘 要:[目的]选择一种较优的黄单胞杆菌EPS提取方法,并分析环境因素对EPS分泌量的影响。[方法]分别采用离心、加热、超声、硫酸、EDTA和甲醛加碱法提取EPS,测定提取物中多糖、蛋白质及DNA含量,以评价提取效率及对细胞的破坏程度。利用正交试验设计考察温度、培养基pH值等因素对EPS分泌量的影响。[结果]甲醛加碱法和超声法的提取效率较高。通过对不同超声功率的比较,选择在50W下超声4min提取EPS。正交试验结果表明,黄单胞杆菌EPS最大分泌量的环境条件为pH值6.0,温度32℃,菌液比7∶100,金属盐溶液投加量3ml。[结论]黄单胞杆菌EPS提取采用超声法;环境因素中温度对分泌量的影响最大。[Objective ] The aim was to compare different mathods of extracting EPS from xanthomonas and study the effect of environmental factors on the yield of EPS.[Method]Six methods including centrifugation,heating,sonication,sulfuric acid,EDTA,formaldehyde plus sodium hydroxide were adopted to extract EPS,and then the extraction abilities of these methods were evaluated in terms of extraction efficiency and cells destruction through measuring concentrations of polysaccharide,protein and DNA respectively.The orthogonal design was applied to study the effect of environ-mental factors on the yield of EPS,such as the pH of medium and temperature.[Result]The result showed that both the formaldehyde plus sodium hydroxide and sonication had a higher extraction efficiency than the other four methods.Through comparing different ultrasonic powers,sonication(50 W,4 min)was determined as the optimal extraction method.The orthogonal design showed that the condition of the maximum EPS yield was pH 6.0,temperature of 32 ℃,7∶100of the inoculated density,the solution of the metal with 3 ml.[Conclusion]Sonication extracting EPS was choosen and temperature had the most significant impact on the yield of EPS.
