摘 要:目的:探讨1α,25-二羟维生素D3[1α,25-(OH)2D3]对体外培养人输卵管上皮细胞活性及细胞维生素D受体(VDR)、钙结合蛋白calbindin-D28k(CaBP-D28k)表达的影响。方法:以不同浓度(0,10-10,10-9,10-8,10-7mol/L)1α,25-(OH)2D3处理体外培养人输卵管壶腹部上皮细胞,分别于不同时间收集细胞。应用溴化四唑蓝比色法检测细胞活性,逆转录聚合酶链反应法、免疫印迹法检测细胞VDR、CaBP-D28k mRNA及其蛋白表达。结果:1α,25-(OH)2D3对体外培养人输卵管上皮细胞增殖无影响。以不同浓度1α,25-(OH)2D3处理细胞12 h,与对照组比较,10-7mol/L 1α,25-(OH)2D3组细胞VDR、CaBP-D28k mRNA表达显著增加(P均<0.05)。以10-7mol/L1α,25-(OH)2D3处理细胞不同时间,与对照组比较,处理6h,12h后细胞VDR、CaBP-D28k mRNA表达显著增加(P均<0.05);处理24h,48h后细胞VDR、CaBP-D28k蛋白表达显著增加(P均<0.05)。结论:在体外培养条件下,1α,25-(OH)2D3不影响人输卵管上皮细胞增殖,1α,25-(OH)2D3可能通过VDR上调人输卵管上皮细胞CaBP-D28k的表达。Objective:To explore the effects of 1α,25-dihydroxyvitamin D3[1α,25-(OH)2D3]on cell survival and the expressions of vitamin D receptor(VDR) and calbindin-D28k(CaBP-D28k) in human tubal epithelial cells in vitro culture.Methods:Human tubal epithelial cells of the ampullary portion in vitro culture were treated with different 1α,25-(OH)2D3 concentrations(0mol/L,10-10mol/L,10-9mol/L,10-8mol/L,10-7mol/L),then were collected after different time,respectively.Cell survival was measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay,the expressions of VDR,CaBP-D28k mRNA and protein were determined by reverse transcription-polymerase chain reaction and Western blot.Results:1α,25-(OH)2D3 did not affect the proliferation of the epithelial cells in vitro culture.After 12 h of the different treatments,compared with the control,the expression levels of VDR and CaBP-D28k mRNA in the 10-7mol/L 1α,25-(OH)2D3 treatment group were increased remarkably(P0.05 or 0.01).After the different time of the 10-7mol/L 1α,25-(OH)2D3 treatment,compared with the control,the expression levels of VDR,CaBP-D28k mRNA in the 6h,12h treatment groups,the expression levels of VDR,CaBP-D28k protein in the 24h,48h treatment groups were significantly higher,respectively(P0.05 or 0.01).Conclusions:1α,25-(OH)2D3 would not affect the proliferation of human tubal epithelial cells in vitro culture.It might up-regulate the expression level of CaBP-D28k through VDR pathway in human tubal epithelial cells in vitro culture.
