摘  要:本研究以东方百合‘索邦'(Lilium Oriental‘Sorbonne')鳞片为外植体获得试管苗,取试管苗鳞片为材料研究细胞分裂素、生长素、培养基、pH值、蔗糖和抗生素等因素对百合试管苗鳞片不定芽分化的影响,建立了东方百合转基因受体系统。结果表明,东方百合试管苗鳞片适宜的再生系统为MS+0.2mg/L6-BA+0.5mg/L2,4-D+20g/L的蔗糖,pH5.8,预培养2d。在抗生素敏感性试验中发现,头孢霉素(Cef)在400mg/L浓度以下对百合试管苗鳞片不定芽分化没有明显影响,但是随着卡那霉素(Km)浓度的升高,不定芽的诱导频率明显降低。本实验还研究了卡那霉素(Km)对试管苗鳞茎生根能力的影响,发现Km在100mg/L以上时能够有效抑制试管苗生根。因此,可以通过在转基因植株的生根培养基中添加一定浓度的Km,来初步检测转基因植株的真伪。In this study,we studied the effects of various factors on adventitious buds induction,such as cytokines,auxin,solid media,pH,sugarand inorganic salts by employing bulb scales of Oriental lily as explants.The results indicated that the best system forsorbonne adventitions bulb induction was Ms added with 0.2 mg/L 6-BA,0.5 mg/L 2,4-D,20 g/L sugarand pre-cultured for2 days.The effect of kanamycin(Km) and cefotaxime(Cef) was also studied.The results indicated that cef with concentration less than 400 mg/L had no effect to adventitious buds induction.But with the rising of Km concentration,the percentage of buds induction was significantly decreased.Effects of the kanamycin to in vitro plantlet rooting was also studied.The results showed that kanamycin could reduce the plantlet rooting efficiently.So it could be used to identify the authenticity of the transgenic plant by adding Km to the culture medium.
