摘　　要：目的观察M3受体激动剂（环戊硫酮）治疗原发性干燥综合征（Primary Sjogren syndrome,pSS）的疗效及安全性。方法对60例pSS进行了环戊硫酮的开放性临床试验,环戊硫酮75 mg.d-1,治疗4w。结果①60例pSS患者为初治患者,平均年龄（44±6.4）岁,平均病程（41.2±5.9）月,男性2例,女性58例。②环戊硫酮能明显改善患者口腔舒适度和咀嚼吞咽能力;19例患者便秘症状消失。③环戊硫酮能明显增加唾液流率[治疗前（1.051±0.619）g/2 min;治疗后（1.324±0.726）g/2 min,P=0.004];泪流率也较治疗前升高[治疗前（4.72±5.67）mm/5 min;（治疗后6.763±5.958）mm/5 min,P=0.034]。④不良反应：治疗组有3例发生与药物有关的不良反应,1例尿频,2例多汗,程度较轻。对肝肾功能、血尿常规、大便隐血等无明显影响。结论环戊硫酮对干燥综合征患者有良好的治疗作用且副作用小。Aim To evaluate the efficacy and safety of M3 receptor agonist in the treatment of patients with primary Sjogren’s syndrome（pSS）.Methods Sixty patients with pSS,females 58 and males 2 with an average age of 44.2 ±6.4 years old,the disease duration of 41.2 months,received anethol trithione,open label,75 mg·d^-1.The total courses were 4 weeks.Global evaluation and subjective re-sponses of patients were assessed by questionnaires with visula analog scales.The patients underwent salivary section,tear film break-up time（BUT） and Schirmer’s test before therapy and 4 weeks after.Results There was a significant increase of the salivary secretion [（1.051 ±0.619） g/2 min;vs（1.324 ±0.726） g/2 min,p =0.004]and Schirmer’s test[（4.72 ±5.67） mm/ 5min;vs（6.763 ±5.958） mm/5 min,p = 0.034]after treatment with anethol trithione as compared with that before untreated.Anethol trithione treatment could improve the global assessment of dry mouth,symptoms associated with dry mouth.The constipation with 19 cases vanished.The adverse reactions were little and tolerable.Conclusion The results of this study suggest that therapy with 75 mg anethol trithione daily is effective,safe and well tolerated for the relief of symptoms in patients with pSS.
