摘 要:目的:本文旨在探讨厚朴对正常和休克时胃肠运动的影响,以期为临床治疗重症感染伴发胃肠运动功能障碍提供新的药物。方法:Wistar雄性大鼠,用药前1W在大鼠的胃、十二指肠、空肠的浆膜面埋置银丝双极电极,随机分为4组(每组1O只):正常对照组经静注和灌胃给与相当量的生理盐水,正常厚朴用药组给与厚朴水煎剂(6.5g.kg,灌胃),单纯休克组经颈静脉给予大肠杆菌0111B4内毒素(15mg.kg^(-1)),休克厚朴用药组在注入内毒素后即刻给予厚朴水煎剂(剂量同上),观察药物对胃肠电的影响。结果:正常厚朴用药组的胃肠电快波的频率和振幅指数均高于正常对照组(P<0.001),且维持3h之久;内毒素休克时胃电快波频率大于5次/min,胃电快波的振幅指数、十二指肠、空肠电的快波频率及振幅指数均明显降低,厚朴水煎剂(剂量同上)使休克时的胃肠电抑制得到明显改善,胃电快波的振幅指数升至26.94±8.39μv/min(单纯休克组:14.88±2.62μv/min,P<0.001),休克用厚朴组及单纯休克组的十二指肠、空肠电的快波频率(次/min)及振幅指数(μv/min)分别为13.08±0.91vs8.89±0.80、100.86±4.96vs68.58±4.76、22.06±0.89vs12.3±1.01、157.22±16.30vs 108.75±8.34,两者比较具有显著性差异,P<0.001。结论:厚朴具有兴奋正常胃肠运动的作用,且明显改善?Aim:To determine the effect of Magnolia bark on gastrointestinal(GI)motility in normal rats and endotoxin shock rats. Methods: Male Wistar rats weighing 250 ~ 300g,were used for this experiments. The GI electrical activity was recorded by three bipolar electrodes implanted chronically on serosa surface of stomach , duodenum and jejunum respectively a week ago. Intravenous injection of endotoxin was given to induce shock .Observe the GI electric activity at 0,30,90, 150min after the adminitration of Magnolia bark by p. o. Compared the difference of the freqnency and amplitude index (AI) of the fast wave among groups with T - test . Result:(l) Magnolia bark(6.5g/kg)increased the electrical activity with the frequency fastened and AI increased,lasted for 2.5 hours or more,compared with control ,P< 0.001. (2) The frequency and AI of GI electrical activity (except the frequency of gastric electrical activity)sharply decreased during endotoxin shock as compared with that of normal rat (P < 0.001) , while the frequency of gastric electrical activity increased. (3) The freqnrncy of gastric electrical activty was not altered by Magnolia bark wllile the AI of gastric electrical activity increased to 26.94±8.39μv/min(that in shock group without drug: 14.88 ± 1.62μv/ min,P< 0.001) .The frequency and AI of duodenal and jejunal electrical activity became higher than that in shock group(drug used group vs control: frequency of duodenum: 13.08 ± 0.91 vs 889 ± 0.80, frequency of jejumum: 100.86 ± 4.96 vs 68.58 ± 4.76, AI of duodenum: 22.06±0.89 vs 12.31 ± 1.01.AI of jejumum :157.22 ± 16.30 vs108.75 ± 8.34,P< 0.001). Conclusion: Magnolia bark may incite the normal GI motility and improve the CI motility during endotoxin shock. It may be a potential prokinetic drug.
