摘 要:建立了LC-MS/MS法测定人血浆中氟康唑的浓度。采用C18色谱柱,以甲磺酸酚妥拉明为内标,流动相为甲醇-2mmol/L乙酸铵溶液(含0.05%甲酸)(60︰40),电喷雾离子化源,正离子多离子反应监测,监测离子为m/z307.2→238.2(氟康唑)和m/z282.2→212.2(甲磺酸酚妥拉明)。氟康唑在0.03~10μg/ml浓度范围内线性关系良好。提取回收率大于90.9%,日内和日间RSD均小于7%。A LC-MS/MS method was established for the determination of fluconazol in human plasma. A C18 column was used with phentolamine mesylate as the internal standard,with methanol-2 mmol/L ammonium acetate solution (containing 0.5% formic acid)(60︰40) as the mobile phase. ESI was applied and operated in positive ion multiple-reaction monitoring mode. The detected ions were m/z 307.2→238.2 (fluconazol) and m/z 282.2→212.2 (phentolamine mesylate). The calibration curve of fluconazdol was linear in the concentration range of 0.03-10 μg/ml. The average extraction recovery was over 90.9%. The intra-and inter-day RSDs were less than 7%.
