摘 要:目的:建立人单核细胞源性泡沫细胞模型;比较高三酰甘油血症个体单核细胞与健康个体单核细胞发生泡沫化的程度。方法:采用密度离心法和玻璃粘附法分离纯化来源于高三酰甘油血症个体和健康个体外周血单核细胞,置于含10%胎牛血清的RPMI1640培养液中培养,加入PMA(100 nmol/L)诱导3 d以分化为巨噬细胞;对照组加培养基,实验组(高三酰甘油血症个体组、健康个体组)加ox-LDL(100mg/L)诱导2 d以分化为泡沫细胞。油红O染色鉴定泡沫细胞形态学特征,镜下计算脂滴占胞浆超过1/3以上的细胞占总细胞数的百分比。结果:油红O染色显示,与ox-LDL共孵育2 d后,细胞中出现大量的红染脂滴颗粒,形成泡沫细胞。高三酰甘油血症个体组红染脂滴占胞浆超过1/3以上的细胞占总细胞数的百分比与健康个体组相比,P<0.05。结论:本实验方法可成功建立泡沫细胞模型;来自高三酰甘油血症个体单核细胞较健康个体单核细胞更容易被诱导形成泡沫细胞,且其泡沫化程度更高。Objective :To establish a human peripheral blood monocyte - derived foam cell model, and compare the difference of the transformation of human peripheral blood monocyte to foam cells in healthy and hypertriglyceridemia cases. Methods:Monocytes were isolated and purified respectively from peripheral blood in healthy and hypertriglyeeridemia eases by density centrifugation and glass adsorptive process. The isolated ceils were induced and stimulated by phorbol 12- myristate 13 -acetate (PMA) at concentration of 100 nmol/L for 3 days to make them transfer to macrophages in RPMI1640 culture medium (including 10% fetal bovine serum) .The macrophages from hypertriglyceridemia group and healthy group were coincubated with ox- LDL at concentration of 100 mg/L for 2 days to make them transfer to foam cells, while the control was only treated with medium. The foam cells were indentiffed by oil red O staining. The degree of foam ceils formation was compared by counting percentage of more than one third of red - stained particals. Results : Oil red O staining showed a great amount of red - stained partieals appeared in co - incubated cells with ox - LDL for 2 days, which indicated the foam cells formation. The degree of foam cells formation from the hypertriglyceridemia group was higher than that of healthy group ( P 〈 0.05). Conclusion:A foam cell model was successfully established in vitro. Monoeytes from hypertriglyceridemia cases could more easily be induced to foam cells and possess higher foam degree than those from healthy cases.
