摘 要:【目的】通过对11批黄精煎剂中5-羟甲基糠醛含量的考察,为黄精的物质基础与质量控制研究奠定基础。【方法】采用煎煮法获得供试品溶液,高效液相色谱(HPLC)法测定其含量。色谱条件:HypersilODS2色谱柱(250mm×4.6mm,5μm),甲醇—体积分数0.05%磷酸溶液(体积比为10:90)为流动相,流速为1.0mL/min,检测波长为280nm。【结果】11批黄精煎剂中5-羟甲基糠醛含量的差别达30倍,在0.0200~2.0000μg(r=0.9997)范围内线性关系良好,平均回收率为100.3%(sR=1.9%,N=6)。【结论】该方法简便、结果准确,为黄精物质基础研究与质量控制提供了一定参考。Objective The content of 5-hydroxymethylfurfural in 11 batches of the decoction of Rhizoma Polygonati was examined, which will supply evidence for the quality control of Rhizoma Polygonati. Methods The sample solution was obtained by decocting. HPLC was applied, and the chromatographic condition was as follows: Hypersil ODS2 chromatographic column (250mm × 4.6mm, 5μm) , methanol- 0.05% phosphoric acid solution ( 10: 90) as a mobile phase, flow rate being 1.0 mL/min, and detection wavelength at 280 nm. Results There were 30 times of differentiation among the content of 5-hydroxymethylfurfural in 11 batches of the decoction of Rhizoma Polygonati. The linear regression for 5-hydroxymethylfurfural was obtained in the range of 0. 0200 - 2. 0000μg (r =0. 9997). The average recovery was 100.3% with RSD being 1.9% (N =6). Conclusion The method is simple and accurate, and can provide a reference for the material base research and quality control of the decoction of Rhizoma Polygonati.
