摘  要:利用PCR-SSCP技术检测安卡鸡、文昌鸡和如皋鸡Δ9-脂肪酸脱氢酶基因的单核苷酸多态性(SNP),并对不同基因型与胸肌脂肪酸含量进行关联分析。结果表明:在外显子6没有检测到SNP突变,在外显子2检测到T101C和C197T 2个SNP突变,共3种基因型,其中A为优势等位基因,其频率在3个鸡种中均大于0.81,该位点在各品种中均处于Hardy-Weinberg平衡(P>0.05)。脂肪酸含量的最小二乘检验表明,AB基因型个体的多不饱和脂肪酸(PUFA)和必需脂肪酸(EFA)含量显著高于AA和BB基因型个体(P<0.05),提示AB型可能对脂肪酸含量有重要的影响。Single nucleotide polymorphisms (SNPs) of Ag-fatty acid desaturatase gene were detected by PCR-SSCP in this study,and correlation between different genotype and breast muscle fatty acid content was analyzed. The results showed that there was no SNP mutation in exon 6,but two SNPs,T101C and C197T in exon 2 with three'genotypes,in which allele A was predominant,its frequency were beyond to 0.81 in three breeds. This site was in Hardy-Weinberg equilibrium in all three chicken breeds (P〉0.05). Association analysis by least squares test showed that PUFA and EFA contents of the chicken with genotype AB were significantly higher than those of the other two genotypes,AA and BB (P〈0.05). It revealed that AB genotype had an important impact on intramuscular fat.
