摘 要:将有机磷农药通用抗体固化在自行制备的纳米磁珠表面,以甲醇、丙酮、pH及包被纳米磁珠的抗体稀释度为主要影响因素,依据均匀设计法和间接竞争酶联免疫反应原理,对纳米磁珠酶联免疫分析(ELISA)方法进行了优化,并开展了纳米磁珠抗体复合物对多种有机磷农药识别作用的比较研究。结果表明,无论是甲醇还是丙酮,抗体稀释度与溶剂互作对半抑制浓度(IC50)的影响最大;与传统ELISA相比,纳米磁珠间接竞争ELISA对毒死蜱、喹硫磷、敌百虫、三唑磷、乙拌磷、伏杀磷、对硫磷、敌敌畏和久效磷均有较好的识别作用,IC50为1.29~6.34μg/mL,比传统ELISA降低了68.3%~95.6%,灵敏度大大提高。The magnetic beads-based enzyme linked immunosorbent assay (mb-ELISA) was established for detection of organophosphorus pesticide.The magnetic bead-antibody complex applied in mb-ELISA was prepared by immobilization a general organophosphorus pesticides (OPs) antibody on the surface of magnetic beads.The optimization of mb-ELISA was found by means of uniform design with organic solvents,pH and the concentration of antibody used for coating magnetic beads as influence factors.The results showed that the interaction between organic solvents and the concentration of antibody had the greatest impact on IC50.Compared with the classical ELISA,the sensitivity of mb-ELISA was greatly enhanced in detecting OPs such as chlorpyrifos,quinalphos,trichlorfon,triazophos disulfoton,phosalone,parathion,dichlorvos and monocrotophos with the IC50 were decreased by 68.3%-95.6%.
