摘  要:目的探讨标本存放时间及温度对PCR检测乙肝病毒DNA(HBV-DNA)结果的影响。方法收集乙型肝炎表面抗原阳性的血清标本64例,采用荧光定量PCR(FQ-PCR)技术对不同时间点及不同温度条件下的同批标本进行HBV-DNA定量检测。结果在室温和2℃~8℃条件下存放7 d、14 d和30 d的血清标本,其HBVDNA定量结果均无显著性差异(P>0.05)。而在-20℃条件下更可长期保存。结论在本室实验条件下,标本存放时间及温度对HBV-DNA的检测结果无明显影响。若资源有限可放宽标本保存条件来较长期的保存所需标本以供临床及科研等所需。Objective To discuss the influence of serum specimen retention time and the temperature to HBV - DNA quota examination result by FQ - PCR, so as to seek for the ideal and convenient method for serum specimen preservation. Methods Three kinds of temperatures ( room temperature,2 ℃ - 8 ℃and - 20℃ ) were selected to preserve serum sam- pies. FQ - PCR was applied to quantitatively determine HBV - DNA loading. Room temperature and 2 ℃ - 8 ℃preserved specimens were examined periodically at 1 st day, 1 st week,2nd week, 1 st month; -20 ℃ preserved specimens were examined at the 3 rd month. Results There were no evident statistical significances at different time and temperature, respectively (P 〉 0.05 ). Serum samples, can be remained long term. Conclusion Serum specimen retention time and the temperature had no effect on HBV - DNA detection under the conditions of our lab and that could make us propose serum specimen conveniently.
