摘  要:通过免疫后攻毒的方法研究了弓形虫代谢分泌抗原(E/SA)免疫后对不同免疫组仔猪机体白细胞介素-2(IL-2)、白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)和肿瘤坏死因子-α(TNF-α)含量的影响.结果表明:免疫后各免疫组仔猪的IL-2、IL-4、IFN-γ含量均不同程度的升高;其中E/SA组IL-2、IL-4和IFN-γ的水平显著升高,与对照组差异显著(P<0.05),其含量分别由免疫前的105.05、105.33、67.50pg.mL-1升高到免疫后的194.03、342.50、296.50pg.mL-1;TNF-α水平变化不显著.试验证实弓形虫代谢分泌抗原疫苗能够促进仔猪IL-2、IL-4、IFN-γ的分泌,并引起宿主抗弓形虫感染的免疫应答.The effects of the Toxoplasma gondii excreted-secreted antigens(E/SA)vaccine on piglet's IL-2,IL-4,IFN-γ and TNF-α were investigated.The piglets were randomly divided into 6 groups with 5 animals in each group,named E/SA group,E/SA+CpG(unemulsified) group,E/SA+CpG group,E/SA+IL-2 group,E/SA+IL-2+CpG group and the control group.The animals were immunized twice at 2 weeks interval and the challenge infection with T.gondii was conducted 2 weeks after the second vaccination.The blood sera were collected before the first immunization,the second immunization,the challenge,1 and 2 weeks after the second immunization,respectively.The levels of IL-2,IL-4,IFN-γ and TNF-α were detected by ELISA.The levels of IL-2,IL-4,IFN-γ in immunized groups increased and the E/SA group increased significantly compared with the control group(P〈0.05),from 105.05,105.33,67.50 pg·mL^-1 to 194.03,342.50,296.50 pg·mL^-1.TNF-α level did not change significantly at all time.These data indicated that Toxoplasma gondii E/SA vaccine can promote the secretion of IL-2,IL-4 and IFN-γ in piglets and cause the anti-T gondii infection immune response.
