摘 要:本研究以半粒小麦无胚种子为材料,提取DNA并进行PCR扩增检测,同时将剩下的半粒小麦有胚种子进行催芽试验.结果表明:从半粒小麦无胚种子和小麦幼叶中都能提取出质量较高的DNA;用λDNA进行浓度检测发现,小麦幼叶提取的DNA浓度高于半粒小麦无胚种子提取的DNA浓度;分别用所提取的小麦DNA作为PCR扩增模板均能够得到理想的特异条带,满足分子检测的需要;催芽试验表明,半粒小麦有胚种子也能够正常生根发芽,可作为染色体分选、原位杂交和繁殖的材料.DNA was extracted from half-grain embryoless wheat seeds and used to conduct PCR amplification tests in this study.In the meantime,the left half-grain embryo seeds were cultured for germination experiments.The results showed that high-quality DNA was able to be extracted from both half-grain embryoless seeds and young leaves.The concentration test with λDNA indicated that DNA concentration from the young leaves was remarkably higher than that from the half-grain embryoless wheat seeds.Both DNAs were used in PCR amplification tests to obtain ideal specific bands,meeting the needs for molecular tests.In addition,the sprouting experiments indicated that the left half-embryo seeds was able to sprout normally and to be used in chromosome sorting,in situ hybridization,and breeding.
