摘 要:目的初步建立我国致病性钩端螺旋体rRNA基因限制性内切酶酶切位点多态性分类体系。方法采用rRNA基因限制性内切酶酶切位点多态性(MRSP)方法对65个血清型75株国内外钩端螺旋体参考株及27株野生株进行分析。结果16SrRNA及23SrRNA基因在HinfⅠ、DdeⅠ的酶切位点上呈多态性,分别存在18种和24种不同的图谱。结合两种方法则有34种不同的图谱。致病性钩端螺旋体间MRSP谱型差别相对较小,遗传距离较近,而它们与非致病性钩端螺旋体的MRSP谱型相差大,遗传距离远。国内主要流行型的MRSP只表现为MRSP1型和2型,非主要流行型的MR-SP型变化较多。大多数野生株与相应的参考株的MRSP型相同,个别具有不同MRSP型的野生株与相应参考株的遗传距离较近。结论rRNA基因限制性内切酶酶切位点多态性(MRSP)可用于钩体分类。Objective To establish an initial system of identification of Leptospira in China by mapped restriction sites polymorphisms of rRNA gene. Methods 75 Leptospira international and domestic reference strains, belonging to 64 serotypes, and 27 field strains were examined using HinfⅠ and DdeⅠ mapped restriction sites polymorphisms (MRSP) of rRNA gene. Results 18 and 24 MRSP profiles were observed in 16S rRNA and 23S rRNA gene, respectively, and the total MRSP profiles were 34. The profiles of pathogenic strains had less difference from each other but more difference from nonpathogenic strains. Only MRSP 2 and 1 were observed among prevalence strains of China, and there were varieties of MRSPs among the non prevalence strains. The profiles of most field strains are in correspondence with reference strains. Conclusions Restriction sites polymorphisms of rRNA gene (MRSP) for Leptospira can be used to classify and identify Leptospira in China and also to study on molecular epidemiology for leptospirosis.
