隔核内注射吗啡对海马痛单位的影响

摘　　要：目的：探讨隔核内阿片肽及相应的受体在隔核影响海马痛单位中的作用。方法；参照Sawyer兔脑图谱，用SN－2型推进器以2μm／s速度将玻璃微电极插人海马P2．5，L3-6，H6．9处引导神经元自发放电；在A3R1H2（外侧隔核）处插入钨丝刺激电极，作刺激隔核用；在A2R2H2及中脑导水管周围灰质（PAG）P9.5R1H-1.5）处分别埋不锈钢微量注射套管，用牙托水配牙托粉固定，作脑室注射用。结果：共引导85个海马痛单位，选取35个对刺激隔核产生抑制效应的海马痛单位，观察隔核内注射阿片受体激动剂吗啡后抑制效应的变化。发现28个痛单位（80％）注药5min后抑制效应开始增强，维持40min；7个痛单位（20％）反应不明显。而静脉注射0．2mg／kg或PAG注射2μg的阿片受体阻断剂纳洛酮后，均能不同程度阻断隔核内注入吗啡增强隔核对海马痛单位的抑制效应（P＜0．005或P＜0.001）。结论：隔核对海马痛单位的调制作用与阿片肽及其相应的受体活动有关；隔核内阿片受体被激活即有明显增强隔核对海马痛单位的抑制作用，其作用传出途径可能通过PAG。To investigate the effect of opioid peptide and opiate receptor of septal nucleus (SN) in modulating the pain units of hippocampus (HPC) in conscious paralytlc rabbits. Metheds: Referring to 'Sawyer's: The rabbit diencephalon in stereotaxic coordinates', glass microelectrodes were put into P2-5, L3-6,H6-9 of the HPC in 2 μm/s by SN-2 to conduct discharges of neurons. Results: Relection of 35 pain units of HPC which amounted to 43- 75 % of pain units was recorded. These 35 pain units of HPC produced mainly inhibitory effect on stlmulating SN, and observing the change of inhibitory effect after activator morphine of opiate recep-tor was microinjected into SM.It was found that, after 5 minutes of morphine injection (2 μg/μ1 ), the inhibito-ry effect of 28 of 35 the observed pain units of HPC start to increase and remained for 40 minutes (80% ); 7 of 35 the observed pain units of HPC had no reaction (20% ). This effect was blocked by intravenous injection (0. 2 mg/kg) and PAG injection (2 μg/2 μl) of antagonist naloxone (P <0.005 or P <0.001). Conclution:Opioid peptide and opiate receptor take part in the inhibltory modulation effect of SN on the pain units of HPC;the activated opiate receptor of SN can increase the inhibitory modulation effect of SN on HPC pain units; and their pathway probably pass through PAG.
