摘 要:目的观察人重组胰岛素样生长因子I(rhIGFI)对STZ诱发的糖尿病大鼠心脏机械功能的作用,并和牛初乳短链胰岛素样生长因子I(BCtIGFI)相比较。方法STZ55mg/kg空腹腹腔注射(IP)制备糖尿病大鼠模型,随机分为四组:DMC组,生理盐水0.1ml/dIP;DML组,BCtIGFI250ng·kg-1·d-1IP;DMH组,BCtIGFI750ng·kg-1·d-1IP;DMrh组,rhIGFI750ng·kg-1·d-1IP;并设正常对照组,每组5只。实验持续6周。心功能用室内导管法测定,观察指标有:室内压最大值(Peak)、舒张末期室内压(EDP)、室内压上升段最大变化率(dp/dt)、室内压下降段最大变化率(-dp/dt)。同时测定尾尖血血糖(BG)、血清果糖胺(FMN)、甘油三酯(TG)、胆固醇(Ch)。结果与正常大鼠相比,DMC组血清FMN、TG明显升高,同时Peak降低41.4%,dp/dt、dp/dt绝对值分别降低40.3%、58.5%,EDP升高〔(2.00±0.62)mmHgvs(-0.44±0.26)mmHg,P<0.01〕。DML、DMH、D?Objective To investigate the effects of recombinant human insulin like growth factor I (rhIGF I) on cardiac function of STZ induced diabetic rats and compare with these of bovine colostrum truncated IGF I (BC tIGF I). Methods Diabetes was induced in male SD rats (180~220g) by intraperitoneal injection (IP) of STZ at a dose of 55mg/kg and animals were divided into four groups: DM C group (n=5), treated with NS 0.1ml/d IP; DM L group (n=5), treated with BC tIGF I 250ng·kg -1 ·d -1 , IP; DM H group (n=5), treated with BC tIGF I 750ng·kg -1 ·d -1 , IP;DM rh group (n=5), treated with rhIGF I 750ng·kg -1 ·d -1 , IP. Therapy lasted 6 weeks. Cardiac functions including heart rate (HR), LV systolic pressure (Peak), LV end diastolic pressure (EDP), rate of LV contraction (dp/dt) and rate of LV relaxation (-dp/dt) were investigated after a catheter was inserted into carotid artery, then into left ventricular. The plasma was separated for analysis of BG、FMN, Ch and TG. Results Compared with normal group, DM C group had a markedly increased FMN and TG, a 41.4% decrease in Peak, a 40.3% decrease in dp/dt, a 58.5% decrease in -dp/dt and an increased EDP (2.00±0.62)mmHg vs (-0.44±0.26)mmHg, P<0.01 , showing impaired cardiac function. The FMN and TG of DM L, DM H and DM rh were obviously lower than those of DM C, but still higher than those of normal group. The indices of myocardial function of DM L and DM rh were similar to those of normal group (P>0.05).Conclusion IGF I improves metabolism and has a beneficial effect on myocardial functions of STZ induced diabetic rats.
