摘　　要：目的探讨二烯丙基三硫(DATS)对急性肺损伤小鼠血清IL-18及肺湿干重比(Wet/Dry,W/D)的影响。方法 90只健康昆明小鼠,随机分为5组:①ALI组:腹腔注射大肠杆菌脂多糖(LPS);②预防组:腹腔注射DATS7d后注射LPS;③治疗组:腹腔注射LPS后30min注射DATS;④DATS组,腹腔注射DATS7d;⑤对照组:腹腔注射等量氯化钠溶液。光镜观察12h点肺组织形态学改变。测定血清中IL-18在2h和6h含量。测定2h和6h肺湿干重比。结果 DATS预防组肺泡间渗出及炎性细胞浸润较ALI组明显减轻。其血清中IL-18含量均明显低于ALI组(P<0.01)。DATS预防组肺W/D与ALI组相比显著下降(P<0.05)。结论预防性应用DATS可减轻LPS诱导的ALI小鼠肺组织炎症反应及肺水肿,并抑制LPS诱导的小鼠血清中炎性细胞因子IL-18的生成。【Objective】 To observe the effect of DATS on the lung W/D ratio and the inflammatory cytokines IL-18 in serum of LPS-induced ALI mice.【Methods】Ninety healthy Kunming mice were randomly divided into five groups：① ALI group：mice were injected with LPS intraperitoneally;② DATS prevention group：mice were injected DATS intraperitoneally for 7 days and then received LPS injection;③ DATS treatment group：mice were injected LPS intraperitoneally and then received DATS injection 30 min later;④ DATS group：mice received DATS injection intraperitoneally for 7 days and without LPS injection;⑤Control group：mice were received the same bulk of sodium chloride injection.The histopathology changes of lung tissues at 12 h were examined under light microscopy.The levels of IL-18 in the serum were detected with ELISA at 2 h and 6 h.The lung W/D ratio in mice was detected at 2 h and 6 h.【Results】 In DATS prevention group,the concentration of IL-18 in the serum decreased significantly at 2 h and 6 h（P 0.01 vs ALI group） while the lung W/D ratio was reduced（P 0.05 vs ALI group）.【Conclusions】 Pretreatment with DATS could alleviate the degree of pulmonary inflammation and pulmonary edema in LPS-induced ALI mice and inhibit the production of IL-18 in the serum in LPS-induced ALI mice.
