摘 要:目的观察中药复方清热颗粒剂对耐环丙沙星大肠埃希氏菌外排泵耐药机制的影响。方法建立急性耐环丙沙星大肠埃希菌腹腔感染兔模型,复方清热颗粒药液灌胃,空白对照组予等体积蒸馏水灌胃。末次给药心脏取血,离心分离出含药血清和空白血清。应用外排泵抑制剂(CCCP)选择出外排泵阳性的耐环丙沙星大肠埃希氏菌,应用琼脂对倍稀释法检测在有含药血清、空白血清、复方清热颗粒、泵抑制剂的情况下环丙沙星对上述7株大肠埃希氏菌的MIC值变化。结果中药复方清热颗粒含药血清不能明显降低环丙沙星对大肠埃希菌的MIC值。结论复方清热颗粒含药血清不能改善大肠埃希氏菌对环丙沙星的耐药情况。Objective:To obseve the effect of compound clearing-heat granules on the mechanism of CIP-resistance Escherichia coli efflux pump. Methods:Rabbit models were created by the acute CIP-resistant Escherichia coli abonomal infection. Those in the model group were given compound clearing-heat granules through stomache,while the normal group were given water in the same way and the same dose. After the last giving drug,the drug serum and blank serum were separated from heart blood. 7 mutants of positive efflux pump CIP-resisitance Escherichia coli were selected by CCCP. The MIC of CIP adding drug serum,blank serum,compound clearing-heat granules and CCCP to those bacterials were measured by the agar dilution method. Results:Compound Clearing-heat Granules serum can not decrease the MIC of CIP to Escherichia coli. Conclusions:The resistance of the efflux pumps with the peudomonas aeruginosa could not be increased by the compound clearing-heat granules.
