摘 要:目的:观察腹腔注射不同剂量长托宁对大鼠空间学习、记忆、大鼠海马组织Bax/Bcl-2蛋白表达和乙酰胆碱酯酶(AChE)活性的影响。方法:9月龄SD大鼠48只,随机分为4组(n=12)。对照组(C组):腹腔注射生理盐水2ml;长托宁0.64mg/kg(P1)组;长托宁1.28mg/kg(P2)组和长托宁2.56mg/kg(P3)组。停药1d后进行Morris水迷宫行为测试。测试结束后立即处死大鼠,取海马组织,进行免疫组织化学染色,测定海马Bax/Bcl-2蛋白的表达;用比色法进行AChE活性测定。结果:水迷宫测试结果:P3组和P2组大鼠大鼠的潜伏期(2-3d)和游泳距离均明显长于P1组和C组(P<0.05)。Bax蛋白、Bcl-2蛋白在P3组和P2组海马内表达及二者的比值均显著高于P1组和C组(P<0.05)。结论:腹腔重复注射较大剂量长托宁对大鼠的空间学习和记忆能力有明显损害,可能与其影响大鼠海马神经元的凋亡和胆碱能系统有关。Objective:To investigate the effect of Penehyclidine hydrochloride on spatial learning and memory and the changes of Bax/bcl-2levels and acetylcholinesterase(AChE)in the hippocampus of aged rats.Methods:Forty-eight 9-month old SD rats weighing 300-350g were randomly divided into four groups(n=12):control group(group C,intraperitoneal injection of normal saline), 0.64,1.28,and 2.56mg/kg Penehyclidine hydrochloride treatment groups(P1,P2,and P3 groups respectively).Twenty-four hours after treatment,all rats underwent Morris water-maze test(MWM test).The hippocampus samples were harvested to determine the expression of Bax and bcl-2 by using immunohistochemical technique,and the activities of AchE in the hippocampus were examined by colorimetric method.Results:Both the latent period and swimming distance in group P3 and P2 were significantly longer than those in group C and P1(all P0.05).The numbers of Bax and Bcl-2 positive cells were significantly more and the ratio of Bax to Bcl-2 was higher in group P3 and P2 than in group C and P1 respectively(all P0.05).The activities of AChE in group P3 and P2 were significantly higher than in group C and P1(all P0.05).Conclusion:Repeated large dosage administration of Penehyclidine hydrochloride can impair spatial learning and memory of rats maybe through its effects on the apoptosis of hippocampal neurons and themodulations on cholinergic system.
