摘 要:目的:观察量子点纳米荧光材料对组织标本抗原特异性标记的效果。方法:分别对胃腺癌组织标本和膀胱癌组织标本中的肿瘤标志物进行量子点荧光标记,观察标记效果,并在实验后1个月内连续观察荧光标记的稳定性,于实验后当天、10d和30d对荧光强度分4个等级进行评价。结果:在蓝光激发下,阳性标记位点发出橙红色荧光,与组织自发绿色荧光对比明显。持续观察30d,阳性标记位点荧光强度逐渐减弱,97.7%的阳性表达标本的荧光强度在10d内保持稳定,标记30d后荧光强度明显减弱,93%的阳性表达标本的荧光仍在可见范围,但对抗原位点的标识已比较模糊。结论:量子点独特的荧光效应能与传统检测方法有效结合,实现对组织标本敏感、长期的标记。作为一种新的检测手段,量子点荧光具有广阔的应用前景。Objective:To explore the effect quantum-dot-fluorescence labeling for specific detection of antigens in the tissue specimen.Methods:The tumor markers were detected by quantum-based-immunofluorescence in the tissue sample of gastric adenocarcinoma and bladder cancer,respectively,and these markers were observed for one month to evaluate the stability of fluorescent labeling.Results:The orange red fluorescence emerged on the positive marker loci under the stimulating of blue light with obviously contrasting to the tissue spontaneous green fluorescence.Although the intensity of fluorescence gradually weakened in one month after the experiment,the fluorescent intensity of positive expression in 97.7% cases remained stable in the first 10days.The positive marker loci were still could be identified at 30days after the experiment,but is already quite fuzzy.Conclusion:The unique fluorescent properties of quantum dots can be effectively combined with the traditional detection methods,and it is a sensitive and stable tag for detection of tissue antigens.As a new means of fluorescence,quantum dots have broad application prospects.
