摘  要:目的:确定雌激素对体外培养的人真皮成纤维细胞增殖和衰老的影响。方法:运用MTT法检测体外培养成纤维细胞增殖活性;脂质过氧化代谢终产物丙二醛(malondialdehyde,MDA)含量与超氧化物歧化酶(superoxidedismutase,SOD)活性分别采用硫代巴比妥酸染色法和黄嘌呤氧化酶法测定。衰老相关-β-半乳糖苷酶(senescence associated-β-galactosidae,SA-β-gal)染色观察成纤维细胞衰老率的变化。结果:在体外培养条件下,10^(-6)mol/L和10^(-7)mol/L雌激素组成纤维细胞数为0.486±0.012和0.475±0.013,高于对照组的0.409±0.013(Ps<0.05)。上清液中MDA含量为(0.208±0.117)nmol/mL和(0.138±0.205)nmol/mL,低于对照组的(0.255±0.110)nmol/mL(Ps<0.05)。SOD浓度为(22.245±1.924)U/mL和(23.547±1.637)U/mL,高于对照组的(18.606±1.803)U/mL(Ps<0.05)。SA-β-gal阳性细胞数为(19.17±0.98/300)个和(22.17±2.27/300)个,低于对照组的(32.00±1.57/300)个(Ps<0.05)。结论:低浓度雌激素可促进成纤维细胞的增殖和抑制衰老相关-β-半乳糖苷酶的表达。Objective: To determine the effect of 17β- estradiol (17β- E2) on the proliferation and senescence of cultured human dermal fibroblasts. Methods: Human dermal fibroblasts were cultured. The cell proliferative capacity was measured by MTT assay and the senescence associated- β- galactosidae ( SA- β- gal) expression was detected by histochemistry staining. The content of malondialdehyde (MDA) and the activity of superoxidedismutase (SOD) in cultured supernatant were detected. Results: Cultured with 10^-6 mol/L and 10-7 mol/L 17β- E2, the number of fibroblasts was 0. 486 ± 0. 012 and 0. 475 ± 0.013, which was hjgher than that in control group (0. 409 ± 0.013), with a P value 〈 0.05. The content of MDA and SOD in supernatant was 0. 208 ± 0.117 nmol/mL and 0.138 ± 0.205 nmol/mL, and 22.245 ± 1.924 U/mL and 23.547 ± 1.637 U/mL, respectively in 17β - E2 group. Compared with control group (0. 255 ± 0.110 U/mL and 18. 606 ± 1. 803U/mL), The content of MDA was lower (P 〈 0.05) and the content of SOD was higher ( P 〈 0.05). The number of SA - β - gal positive cells in 17β - E2 group was 19.17 ± 0.98/300 and 22.17 ± 2.27/300, while the number in control group was 32.00 ± 1.57/300, with a significant difference (P 〈 0.05). Conclusions: Low level of 17β- E2 can stimulate the proliferation of fibroblasts and inhibit the expression of SA - β- gal.
