摘  要:目的探讨骨质疏松时内源性血清二甲基精氨酸的变化。方法 3月龄雌性SD大鼠50只,实验分对照组和骨质疏松模型组。骨质疏质模型组开腹切除双侧卵巢。12周后腹主静脉取血,处死动物后取下肢骨。骨组织常规切片观察模型复制情况;高效液相色谱法检测并分析比较2组大鼠血清中ADMA的含量。结果骨质疏松组骨小梁变细,数目明显减少,排列不整齐,骨小梁间距增宽。对照组骨小梁丰富,宽度适中,分布均匀,排列规则。血清ADMA检测,骨质疏松组含量高于对照组(P<0.05)。结论大鼠去卵巢12周后发生OP,骨质疏松大鼠体内ADMA含量升高。提示体内缺乏雌激素可以导致ADMA升高,进而抑制NOS,减少NO的产生,最终导致骨质疏松的发生。Objective To explore the alteration of osteoporosis endogenous serum ADMA hydrolyzedenzyme in osteoporosis. Methods Fifty 3-month-old female Sprague-Dawley (SD) rats were laparotomized under general anesthesia. A piece of abdominal fat was cut in control group ( SHAM group) , while bilateral ovarieetomy was undergone in osteoporosis model group (OVX group). All animals were executed after 12 weeks. Blood from abdominal major vein was withdrawn and the bones of lower limb were collected. Bone tissues were routinely sectioned for observation of the model and the level of serum ADMA hydrolyzed-enzyme were determined using high performance liquid chromatography (HPLC). Results In OVX group, trabeculars became thinner and their numbers decreased. They displayed irregularly and their interval distance was widened. In SHAM group, trabeculars were abundant with regular distribution and display. The level of serum ADMA in OVX group was higher than that in SHAM group (P 〈 0. 05). Conclusion After 12 weeks of ovariectomy, osteoporosis occurred and the level of ADMA increased in osteoporosis rats. It was suggested that estrogen deficiency could induce the increase of ADMA level, and then inhibit NOS, and further reduce the production of NO and finally result in osteoporosis.
