摘 要:目的探讨抵当汤及分别加入柴胡、桂枝、黄芪3个加味方对S180荷瘤小鼠外周血IL-8、TNF-α的影响。方法小鼠造模后随机分为模型组、抵当汤组(4 g/kg)、抵当汤加黄芪组(5.67 g/kg)、抵当汤加桂枝组(5.5 g/kg)、抵当汤加柴胡组(5 g/kg),每组15只,另取正常小鼠10只作正常对照组。各用药组灌胃给予相应药物,正常对照组和模型组给等体积0.9%NaCl溶液,每日1次,连续10 d。末次给药后1 h于小鼠眶后静脉取血,离心取血清,采用ELISA方法检测IL-8、TNF-α。结果与正常对照组比较,模型组小鼠血清IL-8水平显著升高(P<0.05);与模型组比较,抵当汤可明显降低S180荷瘤小鼠血清IL-8水平(P<0.05),抵当汤加柴胡组、抵当汤加黄芪组、抵当汤加桂枝组均可明显降低S180荷瘤小鼠血清IL-8水平(P<0.05,P<0.01),其中以抵当汤加柴胡组血清IL-8水平降低最显著(P<0.01)。与正常对照组比较,模型组小鼠血清TNF-α水平明显降低(P<0.05);与模型组比较,抵当汤可明显升高S180荷瘤小鼠血清TNF-α水平(P<0.05),抵当汤加柴胡组、加黄芪组、加桂枝组均可明显升高S180荷瘤小鼠血清TNF-α水平(P<0.05,P<0.01),其中以抵当汤加柴胡组血清TNF-α水平升高最显著(P<0.01)。结论抵当汤及抵当汤加味方能升高S180荷瘤小鼠血清TNF-α水平和降低血清IL-8的表达,其中又以抵当汤加柴胡组效果最为显著。Objective To investigate the effects of "Didang Decoction" and its three modified prescriptions, which Bupleurum, Cinnamon Twig, and Astragalus were added respectively, on the levels of IL-8 and TNF-α in peripheral blood of tumor-bearing mouse. Methods Inoculated intraperitoneally tumors for 8 days, tumor- bearing S180 mice were given "Didang Decoction", "Didang Decoction" plus Bupleurum, "Didang Decoction" plus Cinnamon Twig and "Didang Decoction" plus Astragalus by gavage, 1 time a day for total 10 days; 1 hour after the last administration, the blood was drawn from the ophthalmic venous plexus and centrifuged to obtain serum to detect the levels of IL-8 and TNF-α by ELISA. Results "Didang Decoction" could significantly reduce the level of IL-8 in the serum of tumor-bearing S180 mice, showing a significant difference compared with the model group ( P 〈0.05 ) ; in "Didang Decoction" plus Bupleurum group, "Didang Decoction" plus Cinnamon Twig group and "Didang Decoction" plus Astragalus group, the levels of IL-8 in the serum of S180 mice were decreased significantly(P 〈0.05, P 〈0.01 ), of which the level of IL-8 in "Didang Decoction" plus Bupleurum group decreased most significantly( P 〈0.01 ). "Didang Decoction" could significantly increase the level of TNF-α in the serum of tumor-bearing S180 mice, showing a significant difference compared with the model group( P 〈0.05 ) ; in "Didang Decoction" plus Bupleurum group, "Didang Decoction" plus Cinnamon Twig group and "Didang Decoction" plus Astragalus group, the levels of TNF-α in the serum of S180 mice increased significantly( P 〈0.05, P 〈0.01 ), of which the level of TNF-α in "' Didang Decoction" plus Bupleurum group increased most significantly ( P 〈0.01 ). Conclusion "Didang Decoction" and its modified prescriptions can increase the level of TNF-α and decrease the expression of IL-8 in the serum of tumor-bearlng S180 mice, of which " Didang Decoctio
