摘 要:目的:研究红景天甙(salidroside,Sal)对缺氧(2%~3%)状态下兔肺动脉平滑肌细胞(PASMC)增殖、DNA合成、细胞周期和c-fos,c-myc原癌基因表达的影响。方法:应用细胞培养、四唑盐比色试验(MTT)、[3H]-胸腺嘧啶核苷([3H]-TdR)掺入、细胞周期测定和斑点杂交的方法。结果:发现缺氧24h可直接刺激PASMC,使MTT之OD值增加95%(P<005),[3H]-TdR的掺入量增加140%(P<0.01);在缺氧的PASMC培养液中加入Sal(1200μg/mL)可抑制缺氧的这种促增殖作用,PASMC的MTT之OD值与[3H]-TdR掺入量与缺氧组相比显著下降(P<0.05);流式细胞分析显示缺氧24hPASMCG2/M期细胞比例比常氧组明显增多(P<0.01),G0/G1期细胞比例明显减少(P<0.05),与缺氧组相比Sal可增多PASMCG0/G1期细胞比例,减少G2/M期细胞比例(P<0.05);缺氧可促进c-fos,c-myc原癌基因表达,该效应可被Sal所抑制。结论:红景天甙可抑制缺氧所致的PASMC增殖、DNA合成、进入G2/M期的细胞比例和c-fos,c-my?AIM: To investigate the effects of salidroside on the proliferation, DNA synthesis, cell cycle and c- fos , c- myc oncogene expression of rabbit PASMC(pulmonary artery smooth muscle cell) under hypoxia. METHODS:Techniques of cell culture, MTT test, -TdR incorporation, flowcytometric DNA analysis and Dot blot were used. RESULTS: OD value of MTT and -TdR incorporation to PASMC increased significantly by 95% ( P<0.05 ) and 140% ( P<0 01 ) respectively after 24 h hypoxia, and salidroside(1 200 μg/mL) could inhibit these effects induced by hypoxia( P <0 05); Flowcytometric DNA analysis revealed that hypoxia induced significant enhancement of G 2/M phase of PASMC ( P<0 01 ) and decreased G 0/G 1 phase of PASMC( P <0 05); While salidroside could reverse these effects ( P <0 05); hypoxia stimulated c- fos , c- myc oncogene expression of PASMC, and the effect could be inhibited by salidroside. CONCLUSION:Salidroside inhibited the proliferation, the cell proportion of G 2/M phase, DNA synthesis and c- fos , c- myc oncogene expression of PASMC induced by hypoxia.
