摘  要:此研究采用培养的人胚肺成纤维细胞(HEL)和鼠L-929细胞、分别采用琼脂覆盖法和直接接触法对商业用生物心瓣材料的细胞毒性分析比较。计算浸泡前和浸泡10、20、30天周期中材料的细胞增殖抑制指数,并参照国际标准化组织(ISO)要求评定材料细胞毒性。结果显示:在琼脂覆盖法试验中,无论使用HEL或L-929细胞在生物心瓣材料各浸泡阶段均未观察到明显的脱色区和细胞溶解现象,细胞毒性评价为0-Ⅰ级。在直接接触法试验中,HEL或L-929细胞在生物心瓣材料各浸泡阶段均显示出明显长期的细胞毒性。作者认为,商业用生物心瓣材料的细胞毒性试验使用直接接触法更为敏感与准确。Objective:The difference in cytotoxicity of bioprosthetic cardiac valve materials was compared by using agar overlay and direct contact assay methods.Method:The material was crosslinked by glutaral dehyde and stored in 4% formaldehyde.The cells used for testing were human embryonic fibroblasts and mouse L-929 cells.The agar overlay and direct contact assay methods were used and their results were analyzed and compared.The cell proliferative inhibition index(CPII)was calculated for the testing materials being at the periods before,10,20 and 30 days after immerising the testing materials in the cellular suspension fluid.Conclusion:It was concluded that the direct contact method was more sensitive in the detection of cytotoxicity of the material than that of the agar overlay method.It was considered that in testing the cytotoxicity of the prosthetic heart valve material the direct contact method was preferable because it was more sensitive and accurate.
