罗格列酮对慢性心肌缺血大鼠TNF-a、CX43表达的影响

摘　　要：目的:观察罗格列酮对慢性心肌缺血大鼠心肌纤维化的影响。方法:40只雄性健康清洁级Wistar大鼠,随机分为对照组10只、慢性心肌缺血组15只和罗格列酮干预组15只,慢性心肌缺血组和罗格列酮干预组每日给予4%异丙基肾上腺素50 mg皮下注射,对照组皮下注射等量生理盐水,持续6周。2周后罗格列酮干预组每日用罗格列酮(3 mg/kg)灌胃,持续4周,心肌梗死对照组和假手术组每日用等量生理盐水灌胃。免疫组化法测定大鼠心肌TNF-a、CX43表达及HE染色观察心肌纤维化程度。结果:心肌缺血组和罗格列酮干预组心肌纤维化程度及TNF-a、CX43表达明显高于对照组,罗格列酮干预组显著低于慢性心肌缺血组。结论:罗格列酮可以减少心肌缺血大鼠心肌纤维化,改善心肌重塑。Objective： To observe the effect of rosiglitazone on myocardial fibrosis of chronic myocardial ischemia rats.Methods： Forty male healthy depuratory rats were randomly divided into three groups： control group（10 rats）,chronic myocardial ischemia group（15 rats）,and rosiglitazone intervention group（15 rats）. Rats in chronic myocardial ischemia group and rosiglitazone intervention group were injected 50 mg 4% isoproterenol everyday while rats in control group were injected equal normal saline for 6 weeks.2weeks later,rats in rosiglitazone intervention group were lavaged with rosiglitazone（3 mg/kg） ereryday for 4 weeks.Rats in the other groups were lavaged with equal normal saline.Immunohistochemistry staining was used to detect the expressions of TNF-a and CX43 in myocardium of rats and HE staining was used to detect the extent of myocardial fibrosis.Results： The extent of myocardial fibrosis and the expressions of TNF-a and CX43 were obviously strength in rats of chronic myocardial ischemia group and rosiglitazone intervention group than in control group,and rosiglitazone intervention group were obviously lessened than chronic myocardial ischemia group.Conclusion： Rosiglitazone can decrease myocardial fibrosis of chronic myocardial ischemia rats and improve myocard remodeling.
