柴胡三参胶囊对心肌缺血心律失常模型大鼠心肌细胞Ca^(2+)及SERCA2a mRNA表达的影响

摘　　要：目的:研究柴胡三参胶囊对心肌缺血心律失常模型大鼠的防治作用,通过检查指标探讨其可能的作用机制。方法:以结扎大鼠冠状动脉前降支为模型,将60只大鼠分为模型组、假手术组、柴胡三参胶囊小、中、大剂量组和普罗帕酮组,观察大鼠结扎后心电图、心肌细胞Ca2+浓度及SERCA2amRNA表达。结果:柴胡三参胶囊可减少大鼠心肌缺血心律失常的发生,减少心肌细胞内Ca2+超载,增强SERCA2amRNA的表达。结论:柴胡三参胶囊对大鼠心肌缺血心律失常具有明显的保护作用,可能是通过增加SERCA2amRNA活性,从而减少心肌细胞内Ca2+超载。Objective：To investigate the preventive and therapeutic effect of Bupleurum Sanseng Capsule on the rat model of myocardial ischemia and arrhythmia by examining the indicators,to explore its possible effective mechanism.Methods：through ligating the anterior descending branch of coronary artery,60 rats were randomly divided into the model group,the sham operation group,Bupleurum Sanseng Capsule group with small,medium and large dosages,and Propafenone group.The electrocardiogram（ECG）,the level of cardiac cells Ca2＋ and the SERCA2a mRNA expression in ligated rats were observed.Results：Bupleurum Sanseng Capsules can reduce the possibility of myocardial ischemia and arrhythmia in rat model,reduce Ca2＋ overload within myocardial cells,and enhance SERCA2a mRNA expression.Conclusions：Bupleurum Sanseng Capsule has a significant protective effect on myocardial ischemia and arrhythmia,possibly resulting from increasing the activity of SERCA2a mRNA,and reducing the Ca2＋ overload in myocardial cells.
