摘  要:目的:建立利用鸡胚及其尿囊膜来筛选评价透皮促吸收剂的方法。方法:用鸡胚尿囊膜台盼蓝吸收法测定促吸收剂的刺激性,紫外可见分光光度法在595nm处测定台盼蓝吸收量。在鸡胚气室内加入含有不同透皮促吸收剂的苦参碱溶液,用HPLC法测定各时间点尿囊液中苦参碱的含量。结果:不同促吸收剂的刺激性存在显著差异(P<0.05),氮酮浓度与其刺激性存在较好的线性关系,r=0.9873。空白对照组尿囊液的药物浓度与1%氮酮、1%丙二醇、1%二甲亚砜比较都有统计学意义(P<0.05)。曲线下面积(AUC0-240min)分别为(4000.55±192.57),(13129.22±118.42),(8340.58±367.42),(5110.75±127.61)。结论:1%氮酮的促渗效果好,刺激性较小,利用鸡胚模型评价透皮促吸收剂方便可行,值得深入研究。Objective:To establish the method of evaluating the skin penetration enhancer by chick embryo and chicken embryo chorioallantoic membrane.Methods:Irritancy of penetration enhancer was determinated by the chorioallantoic membrane test with trypan blue staining,the spectrophotometer was adopted to measure the absorptive amount of trypan blue at 595 nm.To add matrine solutions with different penetration enhancers into chick embryo air sac,then HPLC was used to assay the drug concentrations.Results:The discrepancy in irritations of various penetration enhancers(P0.05) could be observed.Linear relationships between concentration and irritation of azone solutions were obvious,r=0.9873.Significant differences of drug concentrations existed among groups of physiological saline,azone,propanediol and dimethyl sulfoxide(P0.05),The AUC0-240 min(μg·mL-1·min-1) were(4 000.55±192.57),(13 129.22 ±118.42),(8 340.58±367.42),(5 110.75±127.61),respectively.Conclusion:It was found that the 1% azone was effective and safe penetration for matrine.To estimate the skin penetration enhancer by this method was economical,feasible and worthy of further study.
