摘 要:目的:通过观察葶苈生脉方对充血性心力衰竭(CHF)大鼠血浆AngⅡ、ALD含量的影响和心肌组织AT1mRNA表达的变化,探讨该复方逆转心室重塑的作用机制。方法:采用腹主动脉缩窄法复制CHF大鼠模型,随机分为假手术组、模型对照组、葶苈生脉方高剂量组、葶苈生脉方低剂量组、阳性药心宝丸对照组。测定血浆中AngⅡ、ALD含量,检测心肌组织AT1mRNA的表达。结果:与假手术组比较,模型对照组大鼠血浆AngⅡ、ALD含量明显升高(P<0.01),心肌组织AT1mRNA表达明显升高(P<0.01);与模型对照组比较,各治疗组大鼠血浆AngⅡ、ALD含量均显著降低(P<0.01),心肌组织AT1mRNA表达均明显降低(P<0.01)。结论:葶苈生脉方能有效调控肾素-血管紧张素-醛固酮系统(RAAS),降低心衰大鼠血浆AngⅡ、ALD含量,下调心肌组织AT1mRNA表达,以达到拮抗心力衰竭的作用。Objective:To explore the mechanism by observing the effects of Tingli Shengmai Decoction on expression of AT1mRNA in myocardial tissue of rats with congestive heart failure(CHF).Methods:The CHF animal models were duplicated by the abdomen arteriarctia method,and the rats were randomly devided into sham,model,high-,low-dose Tingli Shengmai Decoction and masculine medicine(Xinbaowan) control group.The AngⅡ,ALD contents in plasma and the AT1mRNA expression in myocardial tissue of each group were detected.Results:Compared with those of sham group,the AngⅡ,ALD contents in plasma and the expression AT1mRNA in myocardial tissue of model group were significantly increased(P0.01).Compared with those of model group,the AngⅡ,ALD contents in plasma and the expression AT1mRNA in myocardial tissue of each treatment group were significantly decreased(P0.01).Conclusion:Tingli Shengmai Decoction can regulate the AngⅡ,ALD contents in plasma and the AT1mRNA expression in myocardial tissue with CHF.
