摘  要:目的研究血小板源性生长因子A(PDGF-A)及其受体(PDGFR-α)在大鼠急性β射线皮肤损伤创面愈合过程中的变化。方法雌性SD大鼠54只,随机分为3组:照射组(n=24),直线加速器产生的β射线(45Gy)单次照射大鼠臀部皮肤20mm×40mm,建立急性深Ⅱ°β射线皮肤损伤动物模型;烫伤组(n=24),以直径为25mm的恒温电烙铁,温度80℃,时间8s,建立大鼠深Ⅱ°热力烫伤动物模型;对照组(n=6),正常大鼠。采用免疫组织化学观察创面出现后0、1、2、3、4、8周各组大鼠创面组织中PDGF-A/PDGFR-α表达。结果照射组大鼠创面出现后1、2、3周创面组织中PDGF-A和PDGFR-α表达均为阳性,0、4、8周表达为弱阳性。烫伤组烫伤后2周创面组织中PDGF-A和PDGFR-α表达均为强阳性,0、1、3周表达为阳性,4、8周表达为弱阳性。对照组大鼠正常皮肤中PDGF-A及PDGFR-α表达为弱阳性。结论急性β射线皮肤损伤创面愈合过程中PDGF-A及其受体PDGFR-α表达减弱,并持续低表达,不能形成峰值,可能是急性β射线皮肤损伤创面难愈合的机制之一。Objective To investigate the dynamic expressions of platelet-derived growth factor-A(PDGF-A)and its receptor alpha(PDGFR-α)in acute β-ray-irradiated skin injury.Methods Fifty-four female SD rats were divided into 3 groups.The model of acute β-ray-irradiated skin injury in rats was replicated with 45 Gy electron accelerator β-ray on the skin(radiation group,n=24).The model of deep second degree burn in rats was established by electrical scald instrument with constant temperature at 80℃ for 8 s and 25mm were regarded as common traumatogenic factors,respectively(burn group,n=24).Six normal rats were served as the normal controls(normal control group,n=6).The expressions of PDGF-A and PDGFR-α protein were detected by histochemistry.Results The expressions of PDGF-A and PDGFR-α in radiation groups were positive(■)1 week,2 weeks and 3 weeks after wound appeared,which were weakly positive(+)0 week,4 weeks and 8 weeks after wound appeared.The expressions of PDGF-A and PDGFR-α of two burn groups were strongly positive(■)2 weeks after burn,which were positive(■)0 week,1 week and 3 weeks after burn and were weakly positive(+)4 weeks and 8 weeks after burn.The expressions of PDGF-A and PDGFR-α in normal control group were weakly positive(+).Conclusion The continuing low expressions of PDGF-A and PDGFR-α may be partially involved in the mechanism for retarded healing of acute β-ray-irradiated skin injury.
