摘  要:目的探讨淫羊藿总黄酮对心力衰竭(心衰)大鼠心肌一氧化氮(NO)的影响。方法雄性SD大鼠100只,随机分为两组:空白组(n=10);模型组(n=90只)皮下注射异丙基肾上腺素2次(170mg/kg),6周后应用超声心动图检测,以左室射血分数(LVEF)≤45%确定造模成功,将模型分为心衰代偿组、心衰失代偿组,再将心衰失代偿组随机分为心衰对照组、美托洛尔组、淫羊藿总黄酮组(大、中、小剂量),18周后使用左心导管进行血流动力学检查,利用硝酸还原酶法检测心肌NO浓度,并取心肌观察病理形态学特征。结果与空白组相比,心衰代偿和心衰对照组心功能指标显著恶化,心肌NO、心室重量指数明显升高(P<0.01);与心衰对照组相比,淫羊藿总黄酮各剂量组血流动力学指标显著好转,心肌NO、心室重量指数明显降低(P<0.01)。结论淫羊藿总黄酮能够改善实验性心力衰竭大鼠心功能,降低心肌NO水平。Objective To explore the effects of total flavonoids of epimedium(TFE)on nitric oxide(NO)in the myocardium of rats with congestive heart failure(CHF).Methods One hundred male SD rats were randomly allocated to isoproterenol(ISO)-induced CHF groups(ISO,n=90)receiving two subcutaneous injections of 170mg/kg ISO and healthy normal group(NOR,n=10)receiving two subcutaneous injections of 0.25ml normal saline.On the 6th week,left ventricular ejection fraction(LVEF)was measured by echocardiogram and the rats with LVEF≤45% were considered as CHF.CHF rats were divided into 2 subgroups of compensation(COMP)and decompensation(DECOMP).The rats of DECOMP were randomly divided into 5 groups of control(CON,NS 0.1ml·kg-1·d-1),metoprolol(MET,metoprolol 8mg·kg-1·d-1),TFE low-dose(TFE-L),TFE medium-dose(TFE-M)and TFE high-dose(TFE-H).On the 18th week,the heart function parameters(LEVF,FS,LVEDD,LVESD)and LVWI were measured after LV morphopathological examination.NO was detected by enzyme immunoassay method.ResultsCompared with group NOR,heart function was significantly deteriorated with the increases in VWI,LVWI,pathological scores and NO in the myocardium(P0.05)in groups of CON and COMP.Compared with groups of CON and COMP,the VWI,LVWI,pathological scores and NO in the myocardium were decreased distinctly(P0.05)and heart function was improved remarkably in groups of TFE and MET.Conclusion TFE can improve heart function and down-regulate NO in the myocardium of rats with CHF.
