摘  要:目的:构建50种分枝杆菌标准菌株的分枝菌酸(MA)指纹图谱库,并对该库指纹图谱进行比较研究。方法:选择50种标准分枝杆菌,经过接种、培养、皂化及酸化,提取到MA。MA衍生化后,采用高效液相色谱法(HPLC)进行分析,得到相应的指纹图谱。依据谱图的峰族数、相对保留时间及相对峰高详细分析与比对其特征性,并对所选92个临床分离株用本法进行鉴定。结果:通过对分枝杆菌MA的色谱分析,构建了50种分枝杆菌标准菌株的MA指纹谱库。图谱可分为3组,即单族峰型、两族峰型、三族峰型。每组分别包括10、23及17种。在92个临床分离株中,有89株的鉴定结果与已知结果一致,一致率为96.74%。结论:本研究所构建的指纹图谱全部具有特征性,对分枝杆菌的种间鉴定具有一定的参考价值。Objective:To construct fingerprinting library of 50 standard strains of mycobacteria by mycolic acid analysis using high performance liquid chromatography(HPLC) and the characteristic thereof.Methods:Mycolic acids were extracted,derivatized and analyzed by the HPLC method after inoculation,culture,saponification and acidification.The mycolic acid fingerprint library was set up.On the basis of HPLC patterns of relative retention time and relative peak height,the characteristic of the fingerprintings was analyzed and compared.Results:The mycolic acid fingerprinting library of 50 standard stains of mycobacteria was constructed by mycolic acid analysis using HPLC.These patterns may be divided into three groups,such as single-,double-and triple-cluster-peak-patterns,each group comprising 10,23 and 17 species,respectively.Fifty strains were successfully typed by the characteristics.Of 92 isolates tested,89 were given species names in concordance with the known names.The concordance rate was 96.74%.Conclusion:All of patterns have characteristics,which have definite values for identification above mycobacterium species.
