摘  要:目的:探讨癌细胞E2F-1因子的表达状态及其与细胞周期的关系。方法:采用免疫荧光组织化学染色法检测8株胃肠道肿瘤细胞系E2F-1转录因子的表达,并采用流式细胞术检测肿瘤细胞周期的变化。结果:依据E2F-1阳性表达的程度不同,将8株肿瘤细胞系分为E2F-1高表达组(包括HGC-27、BGC-823、SGC-7901、HT29)和E2F-1低表达组(包括MKN45、MKN28、SH288、SW-620)。E2F-1高表达组G1/G0期、S期、细胞凋亡比例分别为(33.48±4.61)%、(63.91±5.02)%、(1.50±0.56)%,低表达组分别为(58.58±3.41)%、(35.38±4.03)%、(5.61±1.44)%,2组间比较差异有统计学意义(t分别为6.04、5.99和2.44,P<0.05或P<0.01)。结论:肿瘤细胞E2F-1因子的表达状态对细胞周期有明显的影响。E2F-1阳性的肿瘤细胞则倾向于更高的增殖速率。Objective:To investigate the activity of E2F-1 transcriptional factor in the cell cycle of gastrointestinal cancer cells.Methods:The expression of E2F-1 factor was detected immunohistochemically in 8 gastrointestinal cancer cell lines.The cell cycle was analyzed by flow cytometry in cancer cells.Results:The gastrointestinal cancer cells were divided into two groups,the E2F-1 high-expression group(including HGC-27,BGC-823,SGC-7901 and HT29) and E2F-1 low-expression group(including MKN45,MKN28,SH288 and SW-620).The ratios of G1/G0 phase,S phase and apoptosis were(33.48± 4.61)%,(63.91±5.02)% and(1.50±0.56)% in high-expression group,and(58.58±3.41)%,(35.38±4.03)% and(5.61± 1.44)% in low-expression group.There were differences between these two groups.Conclusion:The status of E2F-1 expression has a great effect on cell cycle in cancer cells.The E2F-1-positive cancer cells tend to a higher proliferation rate.
