摘 要:目的研究姜黄素固体分散体对游离SiO2染尘大鼠血清及肺组织中肿瘤坏死因子(TNF-α)、转化生长因子(TGF-β1)表达的影响。方法 Wistar大鼠随机分为5组,除正常对照组外,其余4组均用游离Si O2染尘。染尘后,除模型组外,其余3组于第2天给予姜黄素(50 mg/kg)、姜黄素固体分散体低、中剂量组(50、100 mg/kg)干预。第28天后,各组大鼠断颈处死,采用酶联免疫法测定肺组织和血清中TNF-α、TGF-β1的含量。结果模型组肺组织匀浆、血清中TGF-β1、TNF-α含量均明显升高(P<0.01)。姜黄素组、姜黄素固体分散体低、中剂量组均可降低肺组织匀浆及血清中TGF-β1、TNF-α的含量(P<0.01),与姜黄素组比较,姜黄素固体分散体组肺组织匀浆及血清中TGF-β1、TNF-α的含量降低(P<0.05)。结论姜黄素固体分散体能降低游离Si O2染尘大鼠肺组织和血清中TNF-α、TGF-β1含量。Objective To explore the effect of solid curcumin dispersion on TNF-α(tumor necrosis factor α)and TGF-β1(transformation growth factor beta 1)in silica exposed rats.Methods The Wistar rats were randomly divided into 5 groups.Besides the normal control group,the rest 4 groups were exposed to free silica.After silica exposure,three of the four groups were administrated with low and medium dosages of curcumin beginning from the day after exposure.After curcumin intervention for 28 days animals were sacrificed.Levels of TNF-α and TGF-β1 were determined by ELISA.Results Compared with the control group,the contents of TNF-α and TGF-β1 in serum and lung increased significantly in the model group.The contents of TNF-α and TGF-β1 in serum and lung decreased significantly in curcumin group,and low and medium dosages solid curcumin dispersion groups.Conclusions Solid curcumin dispersion could significantly decrease the contents of TNF-α and TGF-β1 in serum and lung of silica exposed rats.
