胶艾颗粒剂对正常小鼠免疫调节作用的影响

摘　　要：目的:探讨胶艾颗粒剂的免疫作用及其机制。方法:取健康昆明小鼠80只随机分为正常对照组、胶艾颗粒剂高、中、低剂量组,每组20只。正常对照组给予生理盐水,胶艾颗粒剂高剂量组灌胃给予试药20g/kg,中剂量组给予10g/kg,低剂量组给予5g/kg,连续15d。采用重量法称量小鼠脾脏和胸腺质量,以其质量与体质量之比作为胸腺和脾脏指数。Giemsa染色计数腹腔巨噬细胞(PMΦ)并计算吞噬百分率和吞噬指数。用比色法测定红细胞(RBC)裂解释放的空斑形成细胞(PFC)量。采用温育法计算绵羊RBC(SRBC)致敏小鼠抗体生成能力。观察Giemsa染色后淋巴细胞个数并计算母细胞转化率。结果:胶艾颗粒剂能增加小鼠脾脏和胸腺的重量(高、中剂量组均P<0.01,低剂量组P<0.05);增强小鼠PMΦ的吞噬能力(高、中剂量组均P<0.01,低剂量组P<0.05);提高小鼠溶血值,增加PFC数(高剂量组P<0.01,中、低剂量组均P<0.05);促进SRBC致敏小鼠抗体生成(高剂量组P<0.01,中、低剂量组均P<0.05);能提高小鼠淋巴细胞转化率(高剂量组P<0.01,中、低剂量组均P<0.05)。高剂量组作用与低剂量组比较,差异有统计学意义(P<0.05)。结论:胶艾颗粒剂的免疫增强作用可能与其能使胸腺和脾脏质量增加,PMΦ的吞噬能力增强,淋巴细胞的转化率升高及抗体能力加强等作用有关。objective:To explore the effects and mechanisms of Jiao'ai granule on immunity.Methods:Eighty mice were randomly divided into four groups,namely: the control group,Jiao'ai granule large dosage(20g/kg) group,Jiao'ai granule middle dosage(10g/kg) group and Jiao'ai granule small dosage(5g/kg) group,20 mice in each group.After the control group was taken in normal saline and the Jiao'ai granule groups were taken in various dosage separately by administration by gavage to mice once a day for 15 days,the spleen and thymus were weighed from the mice,peritoneal macrophages(PMΦ),percentage and index of phagocytosis and lymphocyte transformation rate were counted by Giemsa staining,determined plaque forming cells(PFC) by RBC splited and released with chromatometry,and counted the potential of antibody formation by sensitized mice with sheep red blood cell(SRBC) by incubation.Results:Jiao'ai granule could increase the weight of spleen and thymus(the large and middle dosage group all P <0.01,small dosage group P<0.05),could potentiate phagocytosis function of PMΦ(the large and middle dosage group all P<0.01,small dosage group P<0.05),could enhance hemolytic index and increase PFC quantity(the large dosage group P<0.01,middle and small dosage group all P<0.05),could enhance lymphocyte transformation rate(the large dosage group P<0.01,middle and small dosage group all P <0.05) in mice.The effect of Jiao'ai granule in large dosage was more outstanding than other dosage.Conclusion:It was suggested that Jiao'ai granule is of a great value to enhance immunologic defense function and,by strengthening genuine qi and by restoring normal functioning of the body,may consolidate the constitution.
