摘 要:目的观察参莲方汤剂和提取物对高粘血症大鼠血液流变性的影响。方法 50只大鼠随机分为空白对照组、模型组、阳性药组及参莲方汤剂组、参莲提取物组。参莲方汤剂组和提取物组每日分别给予中药水煎剂和提取物水溶液(10mL/kg),阳性药组给予阿司匹林肠溶片的混悬液(10mL/kg)。空白对照组和模型组灌胃蒸馏水,每日1次,连续7d。给药后第8日,除空白对照组外,采用肾上腺素注射加冰浴的方法复制大鼠高粘血症模型,评价、比较参莲方汤剂和提取物对血液流变学指标的作用。结果与空白对照组比较,模型组的全血粘度、血浆粘度、血浆纤维蛋白原含量、血小板聚集率均显著升高,凝血酶原时间明显缩短,红细胞压积无明显变化;内皮素(ET)和血栓素B2(TXB2)含量明显升高,6-酮-前列腺素F1α(6-keto-PGF1α)含量降低,TXB2/6-keto-PGF1α比值增高。与模型组比较,参莲方汤剂和提取物均可明显降低模型大鼠全血粘度、血浆粘度、血浆纤维蛋白原含量、血小板聚集率,延长凝血酶原时间,两种剂型间差异不明显。参莲方汤剂和提取物组可降低ET和TXB2含量,升高6-keto-PGF1α含量,降低TXB2/6-keto-PGF1α比值。对于ET、6-keto-PGF1α、TXB2/6-keto-PGF1α比值的影响,参莲提取物作用优于汤剂组,差异有统计学意义。结论参莲方提取物可明显改善模型大鼠血液流变学指标,在改善血管内皮细胞功能方面优于参莲方汤剂。Objective To investigate the effects of Shenlianfang decoction and extracts on the hemorheology of hyperviscosity rats. Methods Fifty rats were randomly divided into blank control group, model group, positive drug group, Shenlianfang decoction and extract group. Shenlianfang decoction and extract group were given corresponding solution (10 mL/kg). The positve drug group was given aspirin suspension (10 mL/kg). The blank control group and model group were given distilled water once daily for 7 days. The hyperviscosity model was made by injecting NE and ice bath. The effects of Shenlianfang decoction group and extracts group on hemorheology parameters were evaluated and compared. Results Compared with the blank control group, the blood viscosity, plasma viscosity, level of plasma fibrinogen and platelet aggregation rate of the model group were significantly improved, the prothrombin time was significantly shortened, and hematocrit had no change. ET and TXB2 level in serum were significantly improved, 6-keto-PGFla level was decreased, TXB2/6-keto-PGFla ratio was improved. Compared with the model group, the blood viscosity, plasma viscosity, level of plasma fibrinogen and platelet aggregation rate of Shenlianfang decoction group and extracts group were significantly decreased, the prothrombin time was significantly prolonged, but no obvious difference were founded between the decoction and extracts. Shenlianfang decoction group and extracts group both decreased ET and TXB2 level in serum, improved 6-keto-PGF1Q level, decreased TXB2/6-keto-PGFla ratio. Shenlianfang extracts group had better effects on ET level, 6-keto-PGFla level and TXB2/6-keto-PGF1Q ratio than the decoction group. The difference had statistical significance. Conclusion Shenlianfang extracts can significantly improve the hemorheology parameters of model group rats, and has better effects on improving vascular endothelial cell function than the decoction.
