摘  要:目的:探讨毒扁豆碱对老年痴呆大鼠的空间记忆能力的影响,为老年痴呆病人的临床治疗提供新的治疗思路。方法:健康成年雄性大鼠48只,随机分为痴呆组、毒扁豆碱对照组、毒扁豆碱治疗组、空白对照组。运用大鼠穹窿-海马切断术建立痴呆模型,术后第8天开始腹腔注射毒扁豆碱,连续1周,然后各组进行Morris水迷宫测试。结果:空间探索实验结果显示,大鼠各象限游泳距离占总距离百分率和跨越各象限平台相应位置次数占总次数百分率,毒扁豆碱治疗组均有明显提高。定位航行实验提示毒扁豆碱处理后的痴呆大鼠寻找水下平台时间明显缩短。结论:痴呆大鼠经过毒扁豆碱治疗后,空间记忆能力出现明显好转,提示毒扁豆碱对痴呆大鼠具有一定的改善作用。Objective:To investigate of physostigmine on spatial learning and memory of Alzheimer's disease(AD),provide new treatment method of AD.Methods: 48 male SD rats were randomly divided into one of four groups: AD group,physostigmine group,physostigmine +AD group,control group.AD models were established by fornix-fimbria hippocampus to cut off.After the 8 days of AD model established,physostigmine were intraperitoneal injection twice a day for 3 day,and then each groups were test Morris water maze.Results: In spatial explore test,the percentage of swim distance in the quadrant and times of crossing the platform in physostigmine treatment group rats were significantly increased than those in AD group rats(P〈0.05).In the place navigation test,the escape latency of physostigmine group was significantly shorter than that of AD group.Conclusions: Physostigmine treatment could obviously enhanced the learning and memory ability of AD rats,which suggest that physostigmine a potential agent on AD treatment.
