河南汉族人群7个Y-STR基因座单倍型分析

摘　　要：分析河南汉族群体7个Y-STR基因座单倍型及其在法医学应用,利用PCR扩增检测7个Y染色体特异性的STR基因座,并用非变性聚丙烯酰胺凝胶电泳和银染显色技术分析单倍型频率分布.结果显示:在河南汉族102名无关男性个体中7个基因座DYS438、DYS446、DYS391、DYS390、DYS458、DYS534、DYS426分别检出5、8、7、4、8、6、7个等位基因,共检出100种单倍型,其中98种为唯一的,单倍型基因多样性为0.999 612。7个Y-STR基因座具有很高的识别能力,对建立Y染色体STR数据库,研究群体遗传学和进行法医学鉴定有重要意义。To study haplotypes of 7 Y-specific short tandem repeats（STR） of Han population in Henan province and their application to forensic casework,7 Y-STRs had been amplified by PCR and the haplotype diversity of 7 Y-STRs were analyzed by nondenaturing polyacrylamide gel electrophresis followed by silver stain.The results indicated that DYS438,DYS446,DYS391,DYS390,DYS458,DYS534 and DYS426 showed 5,8,7,4,8,6 and 7 alleles,respectively;a total of 100 different haplotypes were identified in 102 unrelated individuals from the Han population in Henan,of which 98 were found in single individuals,the overall haplotypes diversity reached 0.999 612.The 7 Y-STR loci are highly genetic polymorphism and will be very important to the establishment of Y-STR database and study of population genetics and medicolegal expertise.
