129小鼠CgA基因cDNA 5′端部分片段及CgA基因的克隆

摘　　要：通过ＲＴＰＣＲ方法，从１２９小鼠脑组织中克隆到ＣｇＡ基因ｃＤＮＡ５′端部分片段，序列分析结果表明，所克隆到的片段序列与文献中完全一致。以此片段为探针，从１２９小鼠基因组库中筛选获得阳性噬菌体１１，克隆了ＣｇＡ基因，并与文献中的小鼠ＣｇＡ基因限制性内切酶图谱进行了比较分析。A 129 mouse chromogranin A cDNA 5′ fragment was cloned by reverse transcription polymerase chain reaction (RT PCR) method,using 129 mouse brain tissues as st arting materials.The DNA sequence of the cloned RT PCR product was analyzed,and it was shown that the sequence was the same as in the reference.A positive phag e plaque was obtained from a 129 mouse genomic library by using the cloned frag ment as probe. The restriction map of the cloned 129 CgA gene was analyzed and c ompared with that in the reference.
