摘  要:目的:研究神经管缺陷大鼠模型胚胎形态学改变及PLA2、COX-2表达。方法:16只SD雌性大鼠分为2组:第一组为正常对照组,8只,受孕后正常饲养;第二组为糖尿病模型组,8只,受孕后第6天经过尾静脉注射链脲佐菌素(STZ)构建先天性神经管缺陷组大鼠。妊娠12日后取出所有胚胎做HE染色观察胚胎形态学及免疫荧光染色的方法观察PLA2、COX-2表达。结果:(1)HE染色及免疫荧光发现糖尿病胚胎的神经管形态发育明显畸形(2)免疫荧光发现PLA2、COX-2糖尿病胚胎的表达明显降低。结论:PLA2、COX-2基因表达在SD大鼠神经管的发育过程中起着重要的作用。可能对临床治疗神经管畸形提供了一种新方法。Objective:To detect the morphologic change and expression of PLA2 and COX-2in diabetes-induced embryopathy in rats.Method:Two groups of Sprague-Dawley rats were used.First group had 8 normal control rats with normal diet.Second group had 8 rats treated with streptozotocin(STZ)to induce diabetic with congenital neural tube defects in offspring;all yolk sac cells were harvested at day 12 of gestation from Two group.Embryo morphology HE staining and immunohistochemical experiments were performed to observe the PLA2 and COX-2 expression.Results:(1)HE staining and Immunofluorescence showed obvious morphological deformity differences in embryonic development of neural tube in the normal and diabetes rats.(2)Immunofluorescence showed embryonic yolk sac cell prostaglandin enzyme PLA2 and prostaglandin synthase COX-2 expression were decreased in experimental diabetic group.Conclusion:The expression of PLA2 and COX-2 genes was important in the development of neural tube in the SD rats.It could provide a novel method in the clinical therapy of neural tube defect.
