摘  要:目的试验将单细胞光镊拉曼光技术(LTRS)应用于了解活小细胞内部生化变化过程。方法对加入EB病毒上清液的脐血淋巴细胞进行培养,分别在培养后0 h、24 h、48 h、72 h进行单细胞光镊拉曼光检测,并应用数码相机拍照及记录拉曼光谱。结果 EB病毒感染0 h、24 h、48 h、72 h淋巴细胞胞内物质拉曼光谱在785~1 742 cm-1,对上述不同时间检测的4条拉曼光谱曲线比较研究表明:16种胞内物质随着培养时间的延长,72 h内不断增加,其中以1002 cm-1、1064 cm-1、1265 cm-1、1300 cm-1、1443 cm-1、1655 cm-1、1 742 cm-1归属为苯丙氨酸、脂类、蛋白质、酰胺I/脂类等物质增长较快,785 cm-1所代表的核酸增长的幅度不大。结论单细胞光镊拉曼光技术检测可以应用于研究活的脐血淋巴细胞EB病毒转染后的变化和观察小细胞生长变化过程中内部的生化过程。Objective To study the biochemical change process of biological cells by laser tweezers Raman spectroscopy(LTRS)system.Methods The lymphocytes of fetal blood which had been added supernatants of Epstein-Barr virus were cultured,which were examined at 0 hours,24 hours,48 hours,72 hours by LTRS system and photoed by digital camera respectively.Raman spectra were at 785-1 742 cm-1 when Epstein-Barr virus transferred into the lymphocytes at 0 hours,24 hours,48 hours,72 hours.Results The above-1mentioned Raman spectra curves were comparatively studied.16 kinds of intracellular material increased in 72 hours along with the cultured time extended.Raman spectra at 1002 cm-1,1064 cm-1,1265 cm-1,300 cm-1,1443 cm-1,1655 cm-1,1742 cm-1 increased rapidly,which represented phenylalanine,lipids,protein and amide I/lipids,respectively,and Raman spectra at 785 cm-1 represented nucleic acid increased insignificantly.Conclusion LTRS system can be used to examine the changes of fetal blood lymphocytes transfected by Epstein-Barr virus and the biochemical change process of biological cells.
