摘  要:目的探讨大鼠视神经挤压伤后及修复过程中睫状神经营养因子(ciliary neuro-trophic factor,CNTF)在视网膜上表达的变化及复方麝香注射液对其影响。方法 72只健康SD大鼠,随机分为正常对照组、模型对照组、药物治疗组3组,每组24只。模型对照组和药物治疗组大鼠均采用钳夹法建立视神经挤压伤实验模型,药物治疗组大鼠腹腔注射复方麝香注射液,正常对照组和模型对照组大鼠腹腔注射等容量的灭菌注射用水,分别于造模后第5、15、30d随机抽取各组8只大鼠处死,摘取眼球行视网膜中CNTF的免疫组化检测。结果实验证实正常视网膜中存在CNTF的表达;视神经损伤后5d、15d、30dCNTF表达均增高,伤后第5d最高,15d下降,但仍明显高于正常对照组(P<0.05),至30d时其表达量与正常对照组已无显著性差异(P>0.05);药物治疗组大鼠视神经损伤后5d、15d、30dCNTF表达均明显高于模型对照组(P<0.05),至伤后治疗15dCNTF表达显著高于模型对照组(P<0.01)。结论视神经损伤后视网膜中CNTF表达增加,复方麝香注射液能有效地促进视网膜中CNTF较长时间高表达,这可能是其对视网膜神经节细胞轴突损伤后的保护作用的机理之一。Objective To study the effect of Fufang Shexiang Injection(FSI)on the expression of ciliary neurotrophic factor(CNTF)in retina of rats with optic nerve crushing injury.Methods Seventy-two healthy SD rats were equally divided into three groups:normal group,model group,and treatment group.Rats in model group and treatment group were given the crush of retrobulbarly optic nerve to induce optic nerve crushing injury.Immediately after the model was established,the rats in treatment group were given intraperitoneal injection of FSI for 30 days,and the rats in normal group and model group were given isometrical distilled water.Eyeballs of 8 rats from each group were removed for the examination of retinal CNTF expression at day 5,15 and 30 after the modeling.The method of immunohistochemistry was applied to measure the changes of retinal CNTF.Results There existed protein expression of CNTF in the normal retina.The protein expression of CNTF was increased significantly at day 5,15 and 30 after injury.It reached the peak at day 5 and began to decrease at day 15,but was still significantly higher than that in the normal group(P 0.05).Then it decreased continuously at day 30 without significant difference as compared with the normal group(P 0.05).In the treatment group,the protein expression of CNTF was increased much significantly at day 5,15and 30(P 0.05),and it reached peak at day 15(P 0.01).Conclusion The increase of protein expression of retinal CNTF after optic nerve injury may be a self-protection response of the retinal neurons.FSI can promote and prolong the increase of CNTF protein expression in the retina after optic nerve,which is probably one of its protective mechanisms.
